蛋白激酶C抑制剂星形胞菌素对人胃肠道间质瘤细胞株GIST-T1中血管内皮生长因子表达的影响
Effects of staurosporine on the expression of VEGF in human gastrointestinal stromal tumor cell line of GIST-T1
【摘要】 目的探讨蛋白激酶C(PKC)抑制剂星形胞菌素(STS)在体外对人胃肠道间质瘤细胞株GIST-T1中血管内皮生长因子(VEGF)表达的影响。方法体外培养人胃肠道间质瘤细胞株GIST-T1,MTT法检测STS对GIST-T1细胞株的增殖抑制作用,RT-PCR方法检测VEGF mRNA的表达,Western blot检测VEGF蛋白表达水平的变化。结果 STS呈剂量性抑制人胃肠道间质瘤细胞株GIST-T1的增殖;GIST-T1细胞株高水平表达VEGF,STS能显著降低GIST-T1细胞株中VEGF mRNA和蛋白的表达。结论 STS通过抑制PKC的活性,下调GIST-T1细胞株VEGF的表达,从而抑制人胃肠道间质瘤株GIST-T1的增殖。
【Abstract】 【Objective】 To investigate whether PKC inhibitor staurosporine could inhibit the vascular endothelial growth factor(VEGF) expression of human gastrointestinal stromal tumor line GIST-T1.【Methods】 Human gastrointestinal stromal tumor cell line GIST-T1 was cultured in vitro,proliferation inhibitory effect was tested by MTT assay;VEGF mRNA was measured quantitatively by RT-PCR method,expression of VEGF protein in GIST-T1 was measured by Western blot.【Results】 Staurosporine could inhibit the proliferation of GIST-T1 cells,with dose-related effects;VEGF mRNA expressed in high level in GIST-T1 cells,in which,staurosporine significantly decreased the expression of VEGF mRNA and protein.【Conclusion】 Staurosporine could inhibit proliferation and lower the expression of VEGF in human gastrointestinal stromal tumor cell line GIST-T1 by inhibiting the activation of PKC.
