体外受精中皮质颗粒与多精受精的相关性

【摘要】 目的通过观察小鼠及人不同受精状态时皮质颗粒(CG)的分布情况及分级,找出体外受精(IVF)中CG与多精受精的相关性,以期指导IVF的临床工作。方法选择小鼠不同成熟时期及小鼠和人不同受精状态的卵母细胞,经荧光标记后分别观察CG的分布和迁移情况。结果小鼠2PN的卵母细胞中92.2%的CG分布为Ⅲ级,3PN的卵母细胞中8.6%的CG分布为Ⅲ级;人2PN的卵母细胞中100.0%的CG分布为Ⅲ级,3PN的卵母细胞中17.6%的CG分布为Ⅲ级;小鼠卵母细胞成熟后,体外培养时间分别延长到48和60 h,CGⅢ级所占比例分别降至73.3%和20.0%,同时体外受精的3PN率逐渐增加到10.0%和16.7%。结论 3PN卵母细胞中CG释放不完全,因而不能阻止多精受精;过熟或老化的卵母细胞,CGⅢ级所占比例下降,而3PN率上升;证实了小鼠及人IVF中CG与多精受精具有明显的相关性。

【Abstract】 【Objective】 To observe the different fertility status of mouse and human oocyte’s cortical granule(CG) distribution and classification to find out the correlation of CG and polyspermy in the in vitro fertilization(IVF),finally to guide the clinical practice.【Methods】 Different maturity oocytes of mice and different fertilized state oocytes in mouse and human were labeled by fluorescence and observed the CG’s distribution and migration.【Results】 92.2% mice 2PN oocytes CG distribution of grade Ⅲ,8.6% 3PN oocytes CG distribution of grade Ⅲ;100.0% human 2PN oocytes CG distribution of grade Ⅲ,16.7% 3PN oocytesCG distribution of grade Ⅲ;After mouse oocytes matured,extended in vitro culture time to 48 and 60 h,CG Ⅲ grade percentage decreased to 73.3% and 20.0%,while the 3PN rate gradually increased by 10.0% and 16.7%.【Conclusion】 CG release is incomplete in 3PN oocytes,and therefore can not prevent polyspermy;the rate of oocytes CG distribution of grade Ⅲ is decreased in the over and old matured oocytes.It confirmed the obvious relevance between mouse and
