TLR4在益生菌治疗小鼠Hp感染中的作用
Role of toll-like receptor 4 in probiotic treatment of Hp infected mice
【摘要】 目的比较BALB/c小鼠在幽门螺旋杆菌(Hp)感染后和用益生菌治疗后胃黏膜内Toll样受体4(TLR4)、MyD88和IL-1β在基因和蛋白水平上的表达变化,探讨Toll样受体4信号通路在益生菌治疗Hp感染中的作用。方法 60只感染Hp的BALB/c小鼠随机分为3组,第1组不予治疗,第2组予抗生素三联疗法治疗,第3组予乳酸杆菌治疗,另设20只正常小鼠为对照组。取各组小鼠的胃黏膜标本,用RT-PCR和Western blotting的方法半定量检测TLR4、MyD88、IL-1β在基因和蛋白水平上的表达。结果①正常小鼠胃黏膜内无TLR4、MyD88和IL-1β表达。②益生菌治疗组小鼠的TLR4、MyD88和IL-1β均高于抗生素治疗组,但二者均低于Hp感染组。③各组小鼠TLR4与MyD88的表达均呈正相关。结论 TLR4信号通路参与了Hp的致病机制和益生菌治疗Hp的机制,其信号转导均可能是通过MyD88途径。
【Abstract】 【Objective】 To compare expression of TLR4,MyD88,IL-1β on gene and protein level between after Hp infection and after treatment with probiotics in BALB/c mice.To investigate the role of TLR4 signal pathway in probiotic treatment of Hp infection.【Methods】 60 Hp infected BALB/c mice were randomly divided into 3 groups,Group 1 untreated,Group 2 treated by triple therapy of antibiotic,group 3 treated by Lactobacillus,a separate 20 mice were the control group.RT-PCR and Western blotting were used to semi-quantitative detect of TLR4,MyD88,IL-1β on gene and protein level in the gastric mucosa of mice.【Results】 1.There was no expression of TLR4,MyD88,IL-1β in the gastric mucosa of normal mouse.2.Expression of TLR4,MyD88,IL-1β in probiotic treated mice group were higher than the antibiotic treatment group,but both were lower than Hp infection group.【Conclusions】 TLR4 signal pathway is probably involved in the mechanism of probiotic treatment of Hp infection.
