盐酸小檗碱对DSS诱导小鼠结肠炎COX-2表达的作用
The Effect of Berberine Chloride on COX-2 Expression in DSS-induced Colitis in Mice
【摘要】 目的观察盐酸小檗碱对右旋葡聚糖硫酸钠(DSS)诱导小鼠结肠炎的疗效,探讨可能机制。方法 48只雄性BALB/C小鼠饮用8%DSS溶液成模后分成3组,予不同剂量小檗碱治疗,另16只小鼠予蒸馏水,监测体重及DAI,3d和7d后各组随机处死8只。结肠组织HE染色行组织学评分,免疫组化法检测COX-2蛋白表达。结果小檗碱治疗使小鼠体重增加,DAI积分、组织学评分及COX-2染色积分下降,7d与3d相比,各指标差异有统计学意义(P<0.05),同一时间4组之间各指标差异有统计学意义(P<0.01)。结论小檗碱对DSS诱导小鼠结肠炎有效,抑制COX-2蛋白表达可能是其治疗机制。
【Abstract】 Objective To study the therapeutic effects and the underlying mechanisms of berberine chloride on intestinal inflammation in dextran sulphate sodium(DSS)-induced colitis in mice.Method 48 male BALB/C mice were fed with 8% DSS to induce colitis.After that,mice were randomized into three groups with different dose of berberine chloride therapy.16 mice were fed with distilled water.Weights,disease activity index(DAI) were record every day.After 3 days or 1 week after treatments,mice were scarified and histological scores were evaluated by HE staining.COX-2 protein expression in intestinal tissue was measured by immunohistochemistry staining.Results Treatments with berberine chloride significantly prevent weight loss,reduce the DAI,histological scores and the staining scores of COX-2 protein expression.For all above index,significant differences were found between 3 day-and 7 day-treatment(P<0.05),and within each group and for a given time,between groups of treatment(P<0.01).Conclusions Berberine chloride effectively reduces intestinal inflammation in DSS-induced colitis in mice,which partly depends on the inhibition effect of berberine chloride on COX-2 protein expression.
