正交设计优化总蒽醌测定方法

【摘要】 目的:优化紫外分光光度测定保健食品中总蒽醌的条件。方法:采用正交设计,以总蒽醌含量为指标,对实验中涉及的混合酸比例、溶剂用量等七个因素进行考察并优化。结果:各因素对检测结果影响的顺序为:提取溶剂>水洗次数>混合酸比例>混合碱比例>用水量>用酸量>提取时间。结论:本文运用正交设计有效地解决了实验中需要同时考虑多个因素的问题,将新方法应用于实际检验中,RSD均小于3.28%。

【Abstract】 Objective:To explore the optimum testing conditions of the method for the determination of the total content of anthraquinones in health food.Methods: Indicated by the content of total anthraquinones,seven parameters such as the ratio of the mixed acid,the amount of the mixed acid,were investigated and optimized by orthogonal array experimental designs.Results: According to the results of orthogonal test,the influence of the parameters was: The extracting solvent> the scrubbing frequencies> the ratio of the mixed acid> the ratio of the mixed base> the amount of water> the amount of the mixed acid> the extracting time.Conclusion: Through orthogonal array designs,the problem that multi-factors must be investigated simultaneously had been solved effectively.This method had been applied in the determination of several samples like medicinal materials and health food,and the satisfactory result was obtained(RSD<3.28%).
