不同化疗药物介入干预环境下对肝癌细胞系的敏感性

【摘要】 目的检测不同浓度梯度的高浓度化疗药物持续作用不同时间对肝癌细胞系HepG-2的作用,并探索化疗药物有无浓度、时间饱和现象。方法每种化疗药选择5个浓度及四个时间段,采用MTT法分别检测不同浓度及作用时间的8种化疗药物对体外培养的HepG-2细胞的抑制率。结果各种药物不同作用浓度及时间对肝癌细胞系有不同的抑制度;对于任一药物而言,作用浓度—时间之间存在关联,每种药物均有其饱和浓度及作用时间。结论不同化疗药物有其最佳时间—浓度曲线;可以据此优化化疗方案;实现介入治疗个体剂量精确化。

【Abstract】 Objective To detect the role of different high concentration chemotherapy drugs acting with hepatoma cell line HepG-2 for different times of the;and to explore whether if there are saturation concentration and saturation time for each drug and their relations.Methods for each of eightof chemotherapy drug,we selected five concentrations,each concentration acted with HepG-2 by four different time periods,MTT method were used to detected inhibition rate of different combination of concentrations and times of the eightchemotherapy drugs.Results Different combination of concentration and time had different role on HepG-2;for any drugs,different concentration-time combination had certain association,each drugs has its saturation concentration and time.Conclusion Different chemotherapy drugs have their best concentration-time combination curve;this could guide optimize chemotherapy in intervention therapy to achieve precise Individual therapy.
