【摘要】 目的探讨Th17细胞原发性干燥综合征(pSS)患者外周血中表达水平及意义。方法分离pSS患者和健康者外周血单个核细胞(PBMC),磁珠阴选CD4+T细胞,然后加佛波酯/离子霉素,经过固定及透膜处理后进行细胞内染色,流式细胞术(FCM)检测CD4+T细胞内白细胞介素17(IL-17)水平。结果免疫磁珠阴选CD4+T细胞纯度>90%。pSS患者Th17细胞水平(2.14±0.91)显著高于正常对照组(0.56±0.21),P<0.01;pSS患者TH17细胞水平与ANA滴度、Anti-SSA、Anti-SSB、C3、C4无显著相关性(P>0.05)。结论 pSS患者外周血中TH17细胞表达增高,Th17细胞表达异常增高可能是pSS发病的重要环节。更多还原

【Abstract】 Objective To investigate the expression and significance of Th 17 cells from peripheral blood of patients with sjogren’s syndrome(pSS).Methods Intracelluar flow cytometry detection of IL-17 was established using PMA as stimulators after isolution of untouched human CD4+ T cells from PBMC.Results The isolation of untouched human CD4+ T cells from PBMC was effective and its purity was over 90%.The percentage of intracelluar IL-l7 in CD4+ T cells from pSS patients was increased significantly higher.Such percentage in pSS(2.14±0.91) was higher than that of healthy controls(0.56±0.21),P<0.01.There was no correlation between Th 17 cells and serum concentration of Anti-SSA,Anti-SSA,ANA,C3 and C4 in pSS patients(P>0.05).Conclusion There is an abnormal expression of Th 17 cells in pSS patients.These data indicated that Th 17 cells might play certain role in the pathogenesis and progress in pSS.更多还原
