FHIT蛋白及mRNA在食管癌组织中的表达及意义
FHIT protein and mRNA expression in esophageal carcinoma and its clinical significance
【摘要】 目的研究食管癌组织中FHIT蛋白及mRNA的表达变化并探讨其在食管癌发生、发展中的作用。方法采用SP免疫组化检测法和RT-PCR技术检测FHIT蛋白及mRNA在癌组织及相应癌旁组织和正常食管组织中的表达情况。结果 FHIT蛋白在癌组织中缺失表达66%,弱表达32%;与癌旁组织中FHIT蛋白缺失表达18%差异显著(P<0.001)。FHIT mRNA在癌组织、癌旁组织及正常对照组中的缺失率分别为82%、22%、16.7%。随着临床分期越晚,FHIT蛋白和mRNA缺失率呈增高趋势;并且在高分化、中分化和低分化鳞癌中呈逐渐增高趋势。在有淋巴结转移与无淋巴结转移的癌组织中FHIT缺失率的差别有显著意义。结论 FHIT基因与食管癌的发生、发展过程密切相关,其表达减弱或缺失可能增加了食管癌的易感性。FHIT基因的检测在食管癌的早期诊断、治疗以及预后判断等方面具有重要意义。
【Abstract】 Objective To study FHIT protein and mRNA expression in esophageal carcinoma and assay for clinical pathological significance.Methods The expression of FHIT protein was determined by SP immunohistochemistry in 30 cases of normal tissues of the esophagus,and 50 cases of esophageal carcinoma tissues and tumor-adjacent normal tissues by Reverse Transcription Polymerase Chain Reaction and the expression of FHIT mRNA was detected in the same cases.Results The expression of FHIT protein was found absent in 66%of tumor samples and weak in 32%,which was significantly higher than that in tumor-adjacent tissues 18%(P<0.001).In esophageal carcinoma tissues and tumor-adjacent normal tissues and in normal tissues of the esophagus,the rate of FHIT deletion is 82%、22%、16.7% respectively.The absent rates of FHIT protein and mRNA are higher in later clinical stages of esophageal carcinoma.The absent rates of FHIT protein and mRNA in highly differentiated,neutral differentiated and lowly differentiated esophageal carcinoma show a trend of increase.There was significant difference between the two groups:the absent rates of FHIT protein and mRNA in carcinoma of the esophagus with and without local lymph node metastasis.Conclusion It interpreters that the abnormal expression of the FHIT protein and mRNA affects the occurrence of esophageal carcinoma.Its lower or absent expression may increase the incidence of esophageal carcinoma.The detection of the FHIT gene may be an important significance for early diagnosis,treatment,and prediction of esophageal carcinoma.
