红色红曲菌液态发酵产胞外淀粉酶的研究

【摘要】 通过对红曲菌胞外淀粉酶活力的检测初步研究了红色红曲菌液态发酵产淀粉酶的特性。研究表明,红色红曲菌在液态发酵培养过程中分泌的胞外淀粉酶主要为葡萄糖淀粉酶,产酶时间略提前于红曲菌菌丝体生长高峰期,在培养第2天达到最高值。葡萄糖淀粉酶分泌的最适碳源为6%淀粉,最适氮源为2%蛋白胨,发酵最适条件为32℃,初始pH6,接种量5%。红曲菌产葡萄糖淀粉酶的较佳培养条件与红曲菌较适生长条件基本相符。红曲菌在液态发酵过程中几乎不产胞外α-淀粉酶。无论是固态培养还是液态发酵,红色红曲菌的产淀粉酶能力均低于紫色红曲菌。

【Abstract】 The extracellular amylases liquid-fermented by Monascus ruber was investigated and the activity of amylases was determined.The main extracellular amylase secreted by Monascus ruber in liquid-fermentation was glucoamylase,which reached its highest activity at the second day in culture.The growth time is slightly ahead of mycelium growth time.The optimal Carbon source for glucoamylase secretion was 6% starch;the optimal Nitrogen source was 2% peptone.The optimal condition was at 32℃ with initial pH 6,and 5% inoculum.The best culture conditions for glucoamylase production was as similar as the condition for mycelium growth.In the process of liquid-fermentation,α-amylase was not detected.Compared to Monascus purpureus,the production of amylase by Monascus ruber was lower both in solid state culture and in liquid-fermentation.
