人横纹肌样脑膜瘤细胞株的分离和鉴定

【摘要】 目的人横纹肌样脑膜瘤(rhabdoid meningioma,RM)是一种少见的难治性中枢神经系统肿瘤。为建立其体外研究模型,本实验拟从1例确诊的横纹肌样脑膜瘤中分离和鉴定横纹肌样脑膜瘤细胞株。方法利用贴壁培养和肿瘤球培养结合的方法,分离细胞株。利用流式细胞术分析表面抗原标志。用XTT法检测细胞生长活性。用体内移植方法评估其致瘤能力。结果分离培养得到一细胞株,其表面标志为CD44+CD59+CD105+CD11a-CD31-Cd34-,与间质前体细胞具有相似性。其在体外具有长期传代能力,目前已经传了50余代。体内移植试验表明1×105以上细胞具有较一致的颅内致瘤能力,获得的移植瘤与原发人肿瘤具有相似的组织学特征。结论我们首次分离了1例具有间质细胞特征并具有致瘤性的横纹肌样脑膜瘤细胞株,为恶性脑膜瘤的研究提供了一个体外模型。

【Abstract】 Objective Human rhabdoid meningioma(RM) is a rare but intractable tumor.To establish a model for in vitro research,we attempt to establish a cell line from a RM sample.Methods A combination of plastic adherence method,tumor sphere propagation were employed to establish the cell line.The cell surface marker profile,the cell proliferative activity,and the tumorigenic ability of this cell line were examined using flow cytometry analysis,XTT assay,and in vivo experiments respectively.Results The established cell line displayed a CD44+CD59+CD105+CD11a-CD31-CD34-phenotype which was similar to mesenchymal progenitors.The cell line has been passed for more than 50 generations so far.In vivo experiments showed that the RM cell line could successfully establish exnograft tumors in nude mice when injected at great than 1×105 cells intra-cranially.The xenograft tumors exhibited histological features identical to the human original tumor.Conclusions We have successfully established a rhabdoid meningioma cell line with mesenchymal traits and tumorigenic ability from human brain.
