地黄连提取物镇痛抗炎作用及急性毒性

【摘要】 目的:研究地黄连提取物的镇痛抗炎作用,并进行急性毒性试验。方法:小鼠ig 80,40,20 mg.kg-1地黄连提取物1次,采用醋酸致扭体反应、热板法测定痛阈,观察地黄连提取物的镇痛作用;小鼠、大鼠ig 80,40,20 mg.kg-1地黄连提取物,每天1次,连续3 d,抗炎实验采用二甲苯致鼠耳肿胀法、角叉菜胶致大鼠足趾肿胀法和腹腔毛细血管通透性法;采用Bliss法测定了其半数致死量(LD50)。结果:地黄连提取物(80,40 mg.kg-1,qd×1)能减少醋酸所致小鼠扭体次数,明显提高小鼠的痛阈;其提取物80,40,20 mg.kg-1能抑制二甲苯所致小鼠耳廓肿胀、角叉菜胶引起的大鼠足趾肿胀和醋酸所致小鼠腹腔毛细血管通透性增加;其LD50为478.9 mg.kg-1,LD50的95%可信限为(446.8~513.2)mg.kg-1。结论:地黄连提取物对实验动物模型具有显著的镇痛抗炎作用小鼠单次ig的LD50为478.6 mg.kg-1。

【Abstract】 Objective: To study the effect of extract from Munronia henryi Harms(MhH) on anti-inflammation and analgesia and to observe its acute toxicity.Method: Hot-plate procedure and writhing test in mice were adopted in experimental analgesic models,and anti-inflammatory model of mouse ear edema induced by xylene,rat paw edema induced by carrageenin and exudation of abdominal blood capillaries of mice were applied.The median lethal dose(LD50) was determined.Result: MhH(80,40 mg·kg-1,qd×1) could significantly prolong the pain threshold on hot-plate in mice,reduce the writhing times in mice.It could significantly inhibit the ear edema caused by xylene in mice,rat paw edema induced by carrageenin,and exudation of abdominal blood capillaries in mice caused by acetic acid(80,40,20 mg·kg-1,qd×3).The LD50 of MhH was 478.9 mg·kg-1,its 95% confident limit was 446.8-513.2 mg·kg-1.Conclusion: MhH has obvious effect on anti-inflammation and analgesia and its acute toxicity is low.
