克郁舒神颗粒对实验性恶劣心境肝气郁结证大鼠海马BDNF的影响

【摘要】 目的:研究克郁舒神颗粒对实验性恶劣心境肝气郁结证大鼠海马脑源性神经营养因子(BDNF)的影响。方法:选择60只雄性Wistar大鼠,随机分为空白组、模型组、克郁舒神颗粒高、中、低(1.50,0.75,0.375 g.kg-1)剂量组和氟西汀组,每组10只。采用慢性不可预见的温和性应激和孤养方法制备实验性恶劣心境肝气郁结证大鼠模型。应用免疫组织化学法检测大鼠海马BDNF蛋白表达水平。结果:与空白组比较,模型组大鼠大脑海马BDNF阳性神经元细胞明显减少,克郁舒神颗粒中、低剂量可以提高BDNF阳性神经元的面积比,优于高剂量组及氟西汀组(P<0.01);中、低剂量组之间差异无统计学意义。结论:脑组织海马区BDNF含量降低是导致恶劣心境发生的机制之一,克郁舒神颗粒能提高海马BDNF的含量,且疗效优于氟西汀。

【Abstract】 Objective:To observe the effect of Keyu Shushen granule on the brain-derived neurotrophic fac-tor(BDNF)in the hippocampus for experimental dysthymia and stagnation of liver-qi syndrome in rat.Method:Sixty male rats were randomly divided into six groups:control,model,Keyu Shushen granule(1.50,0.75,0.375 g.kg-1),fluoxetine(10 in each group).The rats were put under chronic unpredictable mild stress and separately raised,in order to induce the model of dysthymia.Immunohistochemical method was used to examine the expression of BDNF in the hippocampus of rat.Result:Compared with those in control group,BDNF positive neurons in the hippocampus of rat were decreased obviously in the model group.After treatment,the area ratio of BDNF positive neurons in The Keyu Shushen granule(low dose,middle dose)on experimental dysthymia stagnation of liver-qi syn-drome rat was improved.It was exceeded that of the fluoxetine group(P < 0.01).Difference between middle dose and low dose had no statistical significance.Conclusion:Decrease of the BDNF positive neurons in hippocampus in rat is one of the mechanisms responsible for the dysthymia stagnation disease Keyu Shushen granule can increase thecontent of BDNF in hippocampus with better efficacy compared to fluoxetine.
