高强度聚焦超声结合微泡造影剂杀伤原头蚴的效果
Effect of high intensity focused ultrasound on Echinococcus granulosus protoscolices with the aid of microbubble contrast agent
【摘要】 目的探讨高强度聚焦超声结合不同比例微泡造影剂对离体原头蚴的杀伤效应。方法在原头蚴悬液中加入不同比例的微泡造影剂(含氟脂质体微泡),以相同超声功率及辐照时间作用(治疗声功率为50W,辐照时间为30s)。台盼蓝排斥试验计数原头蚴杀伤率,光镜检测原头蚴形态结构变化。结果超声剂量一定时,加入适宜比例的微泡造影剂组原头蚴杀伤率比单纯超声辐照组高(P<0.05);随着微泡造影剂比例的增大,原头蚴杀伤率呈增高趋势;原头蚴结构损伤严重,台盼蓝染色深,被膜皱缩体积变小,甚至被膜崩解成碎片,小钩及钙颗粒散在分布。结论 HIFU结合微泡造影剂能增强对离体原头蚴的杀伤效应。
【Abstract】 To investigate the protoscolicidal effects of high intensity focused ultrasound(HIFU) with the aid of microbubble contrast agent(MBCA) on the protoscolices of E.granulosus in vitro.The different suspensions of protoscolices mixed with MBCA in different proportions were treated with HIFU under the condition of constant parameter of fixed sound intensity(50 Watt) and treatment time(30 seconds).The protoscolicidal effects of HIFU were investigated by the trypan blue exclusion assay,and the structural changes of protoscolices were observed by microscope.It was found that in the treated groups whose protoscolices suspensions were added in suitable proportions of MBCA,the dead rates of protoscolices treated with HIFU and MBCA were more than those in the groups treated with HIFU only(P<0.05),and the dead rates of protoscolices were related to the dosage of MBCA in a manner of proportion-dependent.It was observed that the protoscolices treated with HIFU and MBCA were damaged,stained deeply,shrinked to smaller sizes,and some dead protoscolices were broken into pieces resulting of the scattered numerous hooklets and calcareous bodies.The trial suggested that the protoscolicidal effect of HIFU can be enhanced by adding of microbubble contrast agent to the hydatid fluids.
