高强度聚焦超声对新生鼠缺血缺氧性脑损伤SYP的影响

【摘要】 目的:研究国家超声重点实验室研制的高强度聚焦超声(Highintensity focused ultrasound,HIFU)对新生鼠缺血缺氧性脑损伤(Hypoxic-ischemic brain damage,HIBD)的突触素(Synaptophysin,SYP)的影响。方法:选取7 d龄SD大鼠192只,随机分成4组:(1)假手术组(n=48,A);(2)模型组(n=48,B);(3)电针组(n=48,C);(4)超声组(n=48,D)。每组又分为治疗1、2、3周3个亚组,每亚组8只,免疫组织化学染色和RT-PCR方法检测脑组织中海马区域SYP表达水平。结果:各时间点电针组和超声组SYP表达明显高于假手术组和模型组(P<0.05)。结论:HIFU穴位治疗可以上调SYP的表达,对HIBD后的脑组织具有神经保护效应。其机制可能是聚焦超声穴位治疗时其物理效应能起到类似针灸时经络传导增加缺血缺氧性脑损伤大鼠脑区突触的作用。

【Abstract】 Objective:To study the treatment of high intensity focused ultrasound(HIFU) in synaptophysin(SYP)of neonatal rats with hypoxic-ischemic brain damage(HIBD).Methods:One hundred and ninety-two 7-day-old SD rats were randomly divided into four groups:1 sham group(n=48,A),2 HIBD model group(n=48,B),3 acupuncture group(n=48,C),4 high intensity focused ultrasound group(n=48,D),every group respectively divided into 1w,2w,3w sub-groups(n=8).The expressions of synaptophysin(SYP)in Hippocampus were examined with immunohistochemical staining and RT-PCR.Results:In high intensity focused ultrasound group and acupuncture group,SYP expression levels were significantly higher than those in sham group and HIBD model group(P<0.05),which had distinct differences compared with the HIBD model group.The expressions of SYP in high intensity focused ultrasound group had no difference compared with acupuncture group.Conclusion:High-intensity focused ultrasound therapy can enhance the expression of SYP level,which has neuroprotective function.Its possible mechanism is that when HIFU is focused on the point,it can play a similar role like acupuncture which can increase the synaptic of HIBD.
