HPLC-ELSD法测定阿卡波糖发酵液中麦芽糖、葡萄糖和阿卡波糖的含量
Determination of maltose,glucose and acarbose in acarbose fermentation broth by HPLC-ELSD
【摘要】 目的建立HPLC-ELSD法测定阿卡波糖发酵液中麦芽糖,葡萄糖和阿卡波糖的含量。方法采用Inertsil氨基柱(4.6mm×250mm,5μm),柱温:30℃;乙腈-水(75:25)为流动相,流速:1.3mL/min;ELSD气化室温度95℃;气体流速:3L/min;气体压力:1.5 105Pa。结果麦芽糖在8~400μg范围内与峰面积有良好的线性关系(r=0.9991),葡萄糖在4~200μg范围内与峰面积有良好的线性关系(r=0.9994),阿卡波糖在1.6~80μg范围内与峰面积有良好的线性关系(r=0.9985),葡萄糖峰,麦芽糖峰和阿卡波糖峰完全分离。结论方法可靠,简便,准确,可用于同时检测阿卡波糖发酵液中底物和产物。
【Abstract】 To establish an HPLC-ELSD method for the determination of maltose,glucose and acarbose in acarbose fermentation broth.Method Column: Inertsil NH2(4.6mm×250mm,5μm),Column temperature: 30℃;Mobile phase: acetonitrile-water(75∶25),flow rate: 1.3mL/min;ELSD temperature of drift tube: 95℃;gas flow: 3L/min;gas pressure: 1.5×105Pa.Results The assay displayed good linearity,the linear range of maltose was 8~400μg(r=0.9991),that of glucose was 4~200μg(r=0.9994) and that of acarbose was 1.6~80μg(r=0.9985).Conclusion The method is simple and can be used for determination of maltose,glucose and acarbose in acarbose fermentation broth.
