重症乙型肝炎患者血清代谢组研究

【摘要】 目的应用核磁共振氢谱(1H-NMR)和主成分分析(PCA)方法研究重症乙肝病人血清的代谢组变化。方法正常人血清8例和重症乙肝患者血清11例,采用BRUKER 500MHz超导核磁共振波谱仪进行检测,找出重症乙肝患者血清代谢组特征,探讨相关生化过程的改变。结果重症乙肝患者和正常人血清的代谢组具有明显差异,前者血清中蛋氨酸、乙酰乙酸盐、三甲胺和葡萄糖含量升高,而乳酸、胆碱和脂的含量降低。结论通过上述代谢物的变化可区分正常人和重症乙肝患者。这种基于核磁共振氢谱和主成分分析的代谢组学方法可为重症乙肝的诊断提供可靠的分子水平上的代谢依据。

【Abstract】 Objective 1H-NMR spectroscopy has been employed to characterize the metabolite composition(i.e.,metabonome) of the serum samples from severe hepatitis B patients in conjunction with principal component analysis(PCA).Methods 1H-NMR sepctra were measured for serum samples from 8 healthy volunteers and 11 severe hepatitis B patients and the spectra data were analyzed by PCA to reveal the metabonomic changes.Results There are clear metabonomic differences between healthy human and severe hepatitis B patients which are highlighted by the increased levels in methionine,acetoacetate,TMAO and glucose together with the decreased levels of lactate,choline and lipids in severe hepatitis B patients.Conclusion The results showed that the metabonomes of serum from severe hepatitis B patients differ markedly from that of the healthy human,which indicated that serum 1H-NMR sepctroscopy in conjunction with PCA offers an efficient tool for understanding the biochemistry of severe hepatitis B at molecular level.
