小青龙汤对哮喘炎性反应的作用及其机制探讨

【摘要】 目的:探讨小青龙汤对哮喘豚鼠免疫炎性反应介质—免疫球蛋白E(IgE)水平和嗜酸性粒细胞浸润的影响。方法:将48只荷兰种豚鼠随机分为正常对照组、模型组、氨茶碱组(50mg/kg)、小青龙汤高、中、低剂量组(4.5g/kg、1.5g/kg、0.5g/kg),每组8只。采用卵清蛋白腹腔注射后雾化吸入建立豚鼠过敏性哮喘模型,通过酶联免疫方法测定血清中IgE的含量,逆转录-链式聚合酶法半定量测定肺组织白细胞介素5(IL-5)基因和巨噬细胞-粒细胞集落刺激因子(GM-CSF)基因表达量。结果:与模型组比较,小青龙汤治疗组可降低血清IgE含量和IL-5、GM-CSF基因的表达量,且差异均有统计学意义(P<0.01,P<0.05)。结论:小青龙汤可通过降低哮喘豚鼠的血IgE含量和IL-5、GM-CSF基因的过度表达,减少肺内EOS的浸润,减轻IgE介导的气道炎症反应。

【Abstract】 Objective: To observe the effects of Xiaoqinglong decoction on the level of IgE that mediates the inflammatory response and the infiltration of inflammatory cells in guinea pigs with asthma.Method: A total of 48 guinea pigs were divided into control group,model group,Aminophylline group,and low dosage(0.5 g/kg),median dosage(1.5 g/kg),and high dosage groups(4.5 g/kg) of Xiaoqinglong decoction.The guinea pigs were sensitized and challenged with ovalbumin to establish an allergic asthma model.The level of serum IgE was measured by ELISA and the expression of IL-5 mRNA and GM-CSF mRNA in the 1ung tissue was assessed by RT-PCR.Results: Compared with model group,different concentrations of Xiaoqinglong decoction treated guinea pigs showed a significant decrease in the level of serum IgE and in the expression of IL-5 mRNA and GM-CSF mRNA in the lung tissue(P<0.01 and P<0.05).Conclusions: Xiaoqinglong decoction can inhibit the IgE-mediated inflammatory response in respiratory system through reducing the level IgE in sera and the expression of IL-5 mRNA and GM-CSF mRNA.
