多西环素作用前后小鼠胸腺上皮细胞蛋白质表达谱的变化

【摘要】 目的:探讨多西环素(doxycycline,Dox)作用前后小鼠胸腺上皮MTEC1细胞系蛋白质表达谱的差异,为阐明多西环素对胸腺细胞作用的机制奠定基础。方法:利用表面加强激光解析电离飞行时间质谱技术(surface enhanced laser desorption/ionization-time of flight-mass spectrometry,SELDI-TOF-MS)分析H4和WCX2芯片捕获的多西环素作用前后差异蛋白表达谱,然后利用蛋白质序列数据库搜索和分析表达改变的蛋白。结果:SELDI-TOF蛋白芯片中H4芯片捕获了13种表达增高蛋白,5种蛋白表达降低。WCX2芯片捕获了14种表达增高蛋白,4种蛋白表达降低。共有5种蛋白在两种芯片中均显示表达增高,其中与细胞增殖和凋亡有关的蛋白为硫氧还原蛋白(thioredoxin,Trx)。结论:多西环素对MTEC1细胞的蛋白表达具有调控作用,有可能促进MTEC1细胞抵抗凋亡。

【Abstract】 Objective: To analyze the proteomic of mouse thymus epithelia cell line treated with or without doxycycline and further illustrates the mechanism of doxycycline action on mouse MTEC1 cells.Methods: Using the surface enhanced laser desorption/ionization time-of-flight mass spectrometry(SELDI-TOF-MS) protein chip to analyse the proteins expression profiling which were captured by H4 protein chip and WCX2 protein chip from mouse MTEC1 cells cultured with or without doxycycline,and to analyze changing proteins by protein database search.Results: There were 13 proteins upregulated and 5 proteins downregulated which captured by H4 protein chip,and 14 proteins upregulated and 4 proteins downregulated which captured by WCX2 protein chip.Furthermore,there were 5 proteins,upregulated by doxycycline,captured by H4,as well as WCX2 protein chip.It was found that an anti-apoptosis related protein thioredoxin was upregulated by doxycycline.Conclusion: Doxycycline can alternate the protein expression of MTEC1,and may contribute to protect the MTEC1 from apoptosis.
