慢性束缚应激大鼠血糖与杏仁核谷氨酸脱羧酶65的相关性

【摘要】 目的:观察慢性束缚应激对大鼠血糖的影响,初步探讨血糖变化与杏仁核谷氨酸脱羧酶65(GAD65)表达变化的联系。方法:每天束缚大鼠3 h,建立束缚应激模型。分别在第7天、14天和21天采集样本,测量血糖值,称量肾上腺,计算肾上腺指数。用免疫组化的方法检测杏仁核GAD65的表达。结果:慢性束缚应激后,大鼠肾上腺指数明显增加(P<0.05)。其中有9只应激大鼠表现出高血糖的症状,同时杏仁核GAD65表达显著增加(P<0.01),而血糖不升高的大鼠GAD65的表达无明显变化。结论:长期慢性束缚应激可使部分大鼠出现高血糖。杏仁核GAD65的表达升高与血糖升高存在相关性。

【Abstract】 Objective: To observe the effect of chronic immobilization stress on blood glucose in rats,and to discuss the relation between stress-induced blood glucose change and expression change of glutamate decarboxylase 65(GAD65) in the amygdala.Method: The rat models of chronic immobilization stress were used in this experiment.They were manacled in the restraint cylinder to limit exercises for 3 hours.Samples were collected at the 7,14 and 21 days respectively,then tested blood glucose levels,calculated the adrenal index,and detected the expression of GAD65 in the amygdala by immunohisto-chemical staining.Result: After stress,adrenal index was significantly increased(P<0.05).The expression of GAD65 was significantly increased in the amygdala(P<0.01) of the 9 rats with hyperglycemia,while it was not significant different among rats without elevated blood glucose.Conclusion: Some rats developed hyperglycemia after chronic stress.There was a correlation between the up-expression of GAD65 in the amygdala and elevated blood glucose levels.
