冠心病患者外周血单个核细胞HCMV-DNA的荧光定量PCR检测
Clinical significance of detection of the HCMV-DNA concentration in PBMCs of patients with coronary heart disease
【摘要】 目的了解冠心病(CHD)患者外周血单个核细胞(PBMCs)人巨细胞病毒(HCMV)-DNA存在情况并探讨其临床意义。方法应用实时荧光定量PCR(real time FQ-PCR)方法,对62例CHD患者,29例其他心血管疾病患者和30例健康体检者的PBMCs内HCMV-DNA进行定量检测。结果 CHD患者PBMCs的HCMV-DNA检出率41.9%,其他心血管疾病组10.3%,正常对照组10.0%,差异有统计学意义(P<0.05)。CHD组HCMV-DNA载量介于103~105拷贝/ml者为12例,占阳性例数的46.1%(12/26),介于105~106拷贝/ml者为8例,占阳性例数的30.7%(8/26)。结论 CHD患者PBMCs的HCMV感染可能在其致动脉粥样硬化性发病过程中发挥重要作用。
【Abstract】 Objective To study the clinical significance of human cytomegalovirus DNA of peripheral blood mononuclear cells(PBMCs) in patients with coronary heart disease(CHD).Methods Subjects were 62 patients with CHD,29 patients with another form of heart disease,and 30 healthy elderly serving as controls.The levels of HCMV-DNA in PBMCs were determined with real-time quantitative PCR.Results Patients with CHD tested positive for HCMV-DNA in PBMCs at a significantly higher rate(41.9%) than did the control group(P<0.05),and 12 patients with CHD(46.1%) had 103-105 copies/ml copies of HCMV-DNA while 8(30.7%) had 105-106 copies/ml of HCMV-DNA.Conclusion Results suggest that HCMV infection of PBMCs in patients with CHD may play an important role in the pathogenesis of atherosclerosis
