男性不育患者精浆活性氧与细胞因子的相关性研究

【摘要】 目的从分子水平探讨男性生殖系感染大肠埃希菌后导致不育的机理,精浆活性氧的含量与肿瘤坏死因子(TNF-α)、白介素-6(IL-6)、白介素-10(IL-10)和白介素18(IL-18)的相互关系及其在不育中的作用。方法选择60例大肠埃希菌感染男性不育患者作为大肠埃希菌感染不育组、30例无感染不育患者和健康已育男性30例分别作为无感染不育对照组和健康对照组,采用硫代巴比妥酸比色法测定精浆丙二醛(MDA)含量,黄嘌呤氧化酶法检测超氧化物歧化酶(SOD)活性,酶联免疫吸附法测定IL-6、IL-10、IL-18和TNF-α的水平,分析MDA与IL-10、IL-18和TNF-α的相关性。结果大肠埃希菌感染男性不育患者精浆MDA含量、IL-6、IL-18和TNF-α含量均高于无感染不育组和健康对照组(P<0.01),IL-10含量和SOD活性下降;精浆MDA含量与TNF-α(r=0.67)和IL-18(r=0.51)呈正相关,与IL-10呈负相关(r=-0.54)。结论生殖系统大肠埃希菌感染所致男性不育与体内活性氧增加、抗氧化物质降低、细胞因子间网络平衡紊乱有关。

【Abstract】 Objective To explore the role of seminal reactive oxygen species and tumor necrosis factor(TNF-α),interleukin-6(IL-6),interleukin-10(IL-10),and interleukin-18(IL-18) in sterile male and explain the molecular mechanism of sterility due to Escherichia coli in men with a genital tract infection.Methods Semen samples were obtained from 30 healthy volunteers(normal group),30 sterile men without infection(sterile men without infection),and 60 sterile men with E.coli infection(men with E.coli infection).The levels of malondialdehyde(MDA) were determined by thiobarbituric acid colorimetry,superoxide dismutase(SOD) activity was determined by xanthine oxidase assay,and IL-6,IL-10,IL-18,and TNF-α were measured with ELISA.The correlation between the levels of ROSes and cytokines in the groups was analyzed statistically.Results Men with E.coli infection had higher levels of MDA,IL-6,IL-18,and TNF-α and lower levels of IL-10 and SOD than did the other two groups of men(P<0.01).MDA was correlated with TNF-α(r=0.67) and IL-18(r=0.51) and inversely correlated with IL-10(r=-0.54).Conclusion The results suggest that increased reactive oxidative stress,a defective ROS scavenging system,and disrupted cytokine balance may lead to sterility in men with E.coli infection.
