铜绿假单胞菌对抗菌药物交叉耐药的体外研究
Cross-resistance of Pseudomonas aeruginosa to antibiotics:a study in vitro
【摘要】 目的体外建立铜绿假单胞菌(PAE)野生株PAEO1耐药菌株,探讨PAEO1对抗菌药物的交叉耐药性。方法采用亚抑菌浓度梯度递增法体外诱导PAEO1对敏感抗菌药物耐药,采用K-B纸片扩散法监测耐药菌株PAEO1对原敏感抗菌药物的敏感性变化。结果头孢哌酮/舒巴坦诱导PAEO1耐药后,PAEO1对亚胺培南/西司他丁、美罗培南、左氧氟沙星和氨曲南敏感性降低,差异有统计学意义(P<0.01);PAEOl经左氧氟沙星诱导耐药后,对环丙沙星,亚胺培南/西司他丁和美罗培南产生耐药,差异有统计学意义(P<0.01);PAEO1经阿米卡星诱导耐药后,对庆大霉素产生耐药,差异有统计学意义(P<0.01),对照组均未发现药敏改变。结论亚抑菌浓度的抗菌药物可诱导野生株PAEO1产生耐药,且所诱导耐药菌株在同类或非同类抗菌药物之间可产生交叉耐药。
【Abstract】 OBJECTIVE To establish the model of Pseudomonas aeruginosa(PAE) drug resistant strain with wild type PAEO1 in vitro,to study the characteristics of cross-resistance to antibiotics of PAE.METHODS PAE drug resistant strain model with wild type PAEO1 by sub-MIC antibacterials of concentration gradient increasing in vitro was established,Kirby-Bauer(K-B) disk diffusion method was used to detect the sensitivity of PAEO1 induced by antibacterials.RESULTS The sensitivity of PAEO1 induced by cefoperazone sodium/sulbactam sodium to imipenem/cilastatin,meropenem,levofloxacin and aztreonam decreased(P<0.001);PAEO1 developed resistance to ciprofloxacin,imipenem/cilastatin and meropenem after indueced by levofloxacin(P<0.001);PAEO1 resisted to amikacin was not sensitive to gentamicin(P<0.001).The sensitivity of PAEO1 in the control group was any disparity(P>0.05).CONCLUSION The sub-MIC environment can induce wild-type P.aeruginosa PAEO1 resistant to antibiotic,and the drug resistant strain can develop cross-resistance in the same kind antibacterials or other kind antibacterials.
