椭圆叶花锚的化学成分研究
Chemical constituents of Halenia elliptica
【摘要】 目的:对龙胆科Gentianaceae花锚属植物椭圆叶花锚Halenia elliptica的干燥全草进行化学成分研究。方法:椭圆叶花锚的干燥全草用90%乙醇提取,依次用石油醚、乙酸乙酯、正丁醇萃取,对乙酸乙酯萃取部分采用各种柱色谱进行分离纯化,通过波谱数据分析(1H-NMR,13C-NMR,MS)进行结构鉴定。结果:从乙酸乙酯萃取部分分离鉴定了12个化合物,其中5个酮,1个裂环烯醚萜苷,以及6个其他类化合物。分别鉴定为8-羟基-2-甲基色原酮(8-hydroxy-2-m ethylchromone,1),5-羟基-2-甲基色原酮(5-m ethoxy-2-m ethylchromone,2),7-表断马钱子苷半缩醛内酯(7-ep i-vogeloside,3),松柏醛(con iferl aldehyde,4),芥子醛(sinapaldehyde,5),norbellid ifolin(6),1-羟基-2,3,4,6-四甲氧基酮(1-hydroxyl-2,3,4,6-tetram ethoxyxanthone,7),1-羟基-2,3,4,7-四甲氧基酮(1-hydroxyl-2,3,4,7-tetram ethoxyxanthone,8),1-羟基-2,3,5-三甲氧基酮(1-hydroxyl-2,3,5-trim ethoxyxanthone,9),壬二酸(azelaic ac id,10),β-谷甾醇(-βsitosterol,11),齐墩果酸(oleanolic ac id,12)。结论:其中化合物1,2为新的天然产物,化合物3~6,10为首次从椭圆叶花锚中分离得到,化合物6显示较好的抗HBsAg和HBeAg活性,其IC50分别为0.77 mmol.L-1和<0.62 mmol.L-1。
【Abstract】 Objective:To study the chemical constituents of Halenia elliptica.Method:The air-dried whole plants of Halenia elliptica were extracted with 90% EtOH.The EtOH extract was condensed to a small amount of volume and extracted with petroleum ether,EtOAc and n-BuOH,successively.The compounds were isolated and purified by column chromatography from the EtOAc fraction,and identified based on spectral analyses(MS,1H-NMR,13C-NMR).Result:12 compounds were isolated from H.elliptica,and characterized as 8-hydroxy-2-methylchromone(1),5-methoxy-2-methylchromone(2),7-epi-vogeloside(3),coniferl aldehyde(4),sinapaldehyde(5),norbellidifolin(6),1-hydroxyl-2,3,4,6-tetramethoxyxanthone(7),1-hydroxyl-2,3,4,7-tetramethoxyxanthone(8),1-hydroxyl-2,3,5-trimethoxyxanthone(9),together with azelaic acid,β-sitosterol,and oleanolic acid.Cloclusion:Compounds 1,2 were new natural compounds and compounds 3-6,10 were obtained from H.elliptica for the first time and compound 6 showed inhibitory activities against HBsAg and HBeAg secretion with IC50 value of 0.77 and<0.62 mmol·L-1,respectively.
