食管癌组织中miRNA表达差异研究

【摘要】 目的探讨食管癌组织microRNA(miRNA)表达的差异,为研究miRNA在食管癌发生、发展中的作用提供新线索。方法 Trizol法抽提食管癌及癌旁组织总RNA,分离miRNA,采用基因芯片技术,将组织中miRNA与哺乳动物miRNA芯片杂交,采用图像软件和SAM version 2.1进行数据分析。结果与正常癌旁组织相比,有23个miRNAs在食管癌组织中有显著差异,包括18个上调和5个下凋。结论部分差异表达miRNAs可能参与食管癌癌变分子机制,为进一步探索提供思路。

【Abstract】 Objective To explore the miRNA(microRNA)differential expression in human esophageal cancer tissue and to provide the evidence that miRNAs are involved in the development of esophagea cancer.Methods Esophageal cancer tissue and side of cancer normal tissue total RNAs were extracted by Trizol,then miRNAs were isolated.by applying the cell miRNAs to mammalian miRNA chip.Data analysis was proceeded by software.Results Total 23 differential expressed miRNAs were difference,including 5 low-expression and 18 over-expression miRNAs.Conclusion Some miRNAs may be involved in the pathogensis of esophagea cancer,which may provide a idea for further study.
