灯盏花素对糖尿病大鼠心肌生长因子β1、转录因子-κB表达的影响
Effects of breviscapine on expressions of TGFβ1、NF-κB in myocardium of diabetic rats
【摘要】 目的:观察灯盏花素(Bre)对链脲佐菌素(STZ)诱导的糖尿病大鼠心肌生长因子(TGFβ1)、转录因子-κB(NF-κB)表达的影响。方法:采用SD大鼠尾静脉注射45mg.kg-1STZ建立实验性糖尿病模型。30只成模大鼠随机分为糖尿病模型组(DM组)、灯盏花素治疗组(Bre-L组,10mg.kg-1.d-1;Bre-H组,20mg.kg-1.d-1),同时设正常对照组(NC组)10只。用药处理8周后检测各组大鼠体质量、血糖、心脏重量指数、心功能,血清LDH、CK含量及心肌组织SOD活性及MDA含量,采用免疫组化方法检测心肌组织中TGFβ1、NF-κB的表达情况。结果:与DM组比较,Bre治疗组大鼠血糖水平无明显变化,但心脏重量指数明显降低,左室内压上升、下降最大速率下降,血清LDH、CK水平及心肌组织MDA含量明显降低,而SOD活性增强,心肌组织TGFβ1、NF-κB的表达显著减少。结论:Bre可明显改善糖尿病大鼠心功能、减轻脂质过氧化损伤和减少心肌TGFβ1、NF-κB表达,对糖尿病大鼠心肌有一定的保护作用。
【Abstract】 OBJECTIVE To investigate the effects of breviscapine on cardiac function and expressions of TGFβ1、NF-κB in myocardium of diabetic rats induced by STZ.METHODS Experimental diabetes model in 30 SD rats was established by injection of STZ(45 mg/kg) through vena caudalis.They were randomly divided into untreated group(DM) and breviscapine groups(Bre).Another 10 rats were set as normal control group.Eight weeks later,blood glucose,cardiac mass index,content of LDH and CK in serum,activity of SOD and contents of MDA in myocardium of rats in all the groups were assayed.Their cardiac function was observed too.Immunohistochemistry method was used to determine the expressions of TGFβ1 and NF-κB in myocardial tissue.RESULTS Compared with DM group,blood gulcose level of rats in Bre group didn’t show an apparent change,whereas cardiac mass index was lowered and cardiac function(±dp/d tmax) was improved obviously.Elevated LDH and CK content in serum and content of MDA in cardiac tissue of diabetic rats were decreased markedly by breviscapine and decreased activity of SOD in myocardium was improved too.At the same time,expressions of TGFβ1 and NF-κB in myocardial tissue of diabetic rats in Bre groups were much less than those in DM group.CONCLUSION Breviscapine has protective effects on myocardium in diabetic rats,which may be associated with lessening expressions of TGFβ1 and NF-κB in myocardial tissue.
