盐酸阿比朵尔胶囊抗甲型H1N1流感病毒的体外实验研究
Inhibitory Effect of Arbidol Hydrochloride Capsule on Influenza Virus A H1N1 in Vitro
【摘要】 目的评价盐酸阿比朵尔原料药、盐酸阿比朵尔胶囊(恩尔欣)、奥司他韦胶囊(达菲)对甲型流感病毒(H1N1)的抑制效率,检验盐酸阿比朵尔胶囊替代奥司他韦胶囊用于流感治疗的可能性。方法本实验以奥司他韦胶囊为阳性对照药物,采用细胞病变效应(CPE)抑制法、噻唑蓝(MTT)比色法和血凝滴度法对其抗甲型H1N1流感病毒的作用进行了观察。结果盐酸阿比朵尔原料药、盐酸阿比朵尔胶囊、奥司他韦胶囊在安全浓度范围内,对100 TC ID50(病毒半数组织感染量)的甲型H1N1流感病毒均有一定抑制作用,且呈明显量-效关系。其中盐酸阿比朵尔原料药抗病毒生物合成作用、对病毒直接杀伤作用和抗病毒吸附、穿入作用三种方式的治疗指数(TI)分别为(3.08±0.34)、(3.55±0.54)、(3.26±0.32);盐酸阿比朵尔胶囊的分别为(3.12±0.40)、(3.41±0.23)、(4.20±0.43);奥司他韦胶囊的依次为(5.34±0.26)、(1.19±0.16)、(1.01±0.38)。结论盐酸阿比朵尔原料药、盐酸阿比朵尔胶囊在阻断流感病毒吸附、穿入作用和直接杀伤作用方面较奥司他韦胶囊效果好(P<0.01)。奥司他韦胶囊的抗病毒生物合成作用较盐酸阿比朵尔和盐酸阿比朵尔胶囊强(P<0.01),因此盐酸阿比朵尔胶囊可以替代奥司他韦胶囊用于预防甲型H1N1流感病毒感染。
【Abstract】 OBJECTIVE To investigate and compare the antiviral activity of arbidol hydrochloride,arbidol hydrochloride capsule and oseltamivir capsule(Tamiflu) against influenza virus A(H1N1) in vitro.METHODS MTT colorimetric assay and hemagglutination titer assay was used to evaluate the antiviral activity of the drugs.Oseltamivir capsule was used as a positive control.RESULTS Arbidol,arbidol capsule and oseltamivir capsule prevented the cytopathic effect of influenza virus A(H1N1),and exhibited a significant dose-effect relationship.When drugs were added before,during or after viral infection,arbidol treatment index(TI) were(3.08±0.34),(3.55±0.54),(3.26±0.32) respectively,while arbidol capsule TIS were(3.12 ± 0.40) and(3.41±0.23),(4.20±0.43) and the TIS of Oseltamivir capsule were(5.34±0.26),(1.19±0.16) and(1.01±0.38).CONCLUSION In vitro infection with influenza virus A(H1N1) can be effectively treated by arbidol,arbidol capsule and oseltamivir capsule.Arbidol and arbidol capsule were effective in directly killing virus and inhibiting viral absorption and penetration,while oseltamivir capsule is more significant in inhibiting viral biosynthesis.Arbidol capsule can substitute oseltamivir capsule for preventing against Influenza virus A(H1N1) infection.
