【摘要】 目的建立川贝母DNA指纹鉴别方法。方法采用CTAB法从川贝母等样品中提取基因组DNA,对其5s rDNA区序列进行扩增并在此基础上展开了分子鉴定研究。经Primer Primer5.0引物设计软件设计川贝母的特异性引物,利用PCR技术对川贝母与其他品种的贝母进行鉴别比较。结果川贝母能扩增出274bp大小的片段,而其他贝母未能扩增出相应片段。结论川贝母DNA具有特异性指纹特征,可作为川贝母与其伪品鉴定的有效方法,该方法简便快速、结果可靠。更多还原

【Abstract】 OBJECTIVE To establish DNA fingerprint differential method for the identification of Fritillaria cirrhosa D.Don.METHODS The CTAB method was used to extract genome DNA from the Fritillaria cirrhosa D.Don.A high hereditary 5s rDNA sequence in GenBank was obtained.The specific primer of Fritillaria cirrhosa D.Don using was designed by software Primer Primer 5.0,and Fritillaria cirrhosa D.Don was identified by PCR(Polymerase Chain Reaction).RESULTS 19 Kb DNA fragments were obtained.Only Fritillaria cirrhosa D.Don(F.prezwalskii Maxim,F.unibracteata Hsiao et K.C.Hsia) showed specificity binding site with primer after the PCR,and its fragment was about 274 bp.CONCLUSION Fritillaria cirrhosa D.Don DNA fingerprint with specific characteristics,it can be used to identify Fritillaria cirrhosa D.Don.The method is effective,simple,rapid and reliable.更多还原
