狭基线纹香茶菜水提部位对免疫肝损伤小鼠NF-κB蛋白表达水平及核转位的影响

【摘要】 目的:研究狭基线纹香茶菜水提部位对免疫肝损伤小鼠核因子κB(NF-κB)蛋白表达水平及核转位的影响。方法:小鼠尾静脉注射刀豆蛋白A(ConA)25mg·kg-1复制肝损伤模型,免疫组化法检测NF-κB在肝组织的表达,以及NF-κB从细胞浆向细胞核转位的情况。结果:复制模型4、8h后小鼠NF-κB蛋白表达显著增加(P<0.01),吡咯烷二硫代氨基甲酸盐(PDTC)和狭基线纹香茶菜水提部位9.2g·kg-1在4、8h均可显著降低模型小鼠NF-κB的蛋白表达量(P<0.01)。ConA能显著促进NF-κB自细胞浆向细胞核转位(P<0.01)。PDTC和狭基线纹香茶菜提取物9.2g·kg-1能显著减少NF-κB在细胞核表达,使其在细胞浆表达显著增多(P<0.01)。结论:狭基线纹香茶菜可抑制免疫肝损伤小鼠NF-κB蛋白的表达,并抑制NF-κB的核转位。狭基线纹香茶菜对抗免疫肝损伤的作用机制可能与其抑制核NF-κB的激活有关。

【Abstract】 OBJECTIVE:To study the effect of water-soluble extract from Isodon lophanthoides on the expression of NF-κB protein and nuclear translocation in mice with immune hepatic injury.METHODS:Hepatic injury model was induced by tail intravenous injection of Con A 25 mg·kg-1.The expression of NF-κB protein and nuclear translocation in liver tissue were detected by immunohistochemistry examination.RESULTS:The expression of NF-κB protein increased significantly 4 h and 8 h after modeling(P<0.01).PDTC and water-soluble extract (9.2 g·kg-1) can inhibit the expression of NF-κB protein(P<0.01).ConA can improve that NF-κB translocated from cytoplasm into nucleus(P<0.01).PDTC and water-soluble extract (9.2 g·kg-1) reduced the expression of NF-κB protein in nucleus,and increased that in cytoplasm.CONCLUSION:I.lophanthoides can down-regulate the expression of NF-κB protein and inhibit nuclear translocation.Inhibiting effect of I.lophanthoides on immune hepatic injury is associated with the inhibition of the activation of NF-κB.
