131I标记大肠癌单抗片段的比活度对细胞结合率的影响

【摘要】 目的探讨不同比活度的131I标记大肠癌单抗片段对大肠癌细胞结合率的影响。方法应用37MBq的131I分别标记1000、200、100、50、25μg的单抗片段,测定标记率>95%后与2×106的大肠癌细胞反应30min,离心、洗涤2次后测量结合率;对照组设骨髓瘤细胞,观察非特异性结合率;选取结合率最高组,增加细胞数,观察结合率变化。结果 1000、200、100、50、25μg的131I大肠癌单抗片段的放射性比活度分别为37、185、370、740、1480kBq/μg;与2×106大肠癌细胞的结合率分别为10.1%、14.7%、35.1%、32.7%、22.6%;非特异性结合率为0.8%;增加细胞数可以进一步增加结合率。结论比活度为370740 kBq/μg的碘标记大肠癌单抗片段对大肠癌细胞有较高的结合率。

【Abstract】 Objective To explore effects of different specific activities of anti-colorectal carcinoma monoclonal antibody fragment labeled by 131I on binding rate of colorectal carcinoma cells.Methods The 37 MBq 131I was adopted to label 1000,200,100,50 and 25μg monoclonal antibody fragment respectively,which reacted with colorectal carcinoma cells for 30 mins after the labeling rate was determined to be greater than 95%.Binding rate was measured after centrifugation and washing for 2 times.The control group of myeloma cells was set up to observe non-specific binding rate.In a group with the highest binding rate,cells were increased to observe the binding rate.Results The specific activity of anti-colorectal carcinoma monoclonal antibody fragment labeled by 1000,200,100,50 and 25μg 131I was 37,185,370,740 and 1480kBq/μg respectively;the binding rate with 2×106 colorectal carcinoma cells was 10.1%,14.7%,35.1%,32.7%and 22.6%respectively;and the non-specific binding rate was 0.8%.Binding rates could be further improved by increasing the cell number.Conclusion Anti-colorectal carcinoma monoclonal antibody fragment with specific activity of 370-740 kBq/μg labeled by iodine had high binding rate with colorectal carcinoma cells.
