高效液相色谱法测定桔霉素

【摘要】 桔霉素是红曲霉的代谢产物,有显著地肾脏毒性。文章研究了HPLC法测定桔霉素,结果表明:色谱柱SHIMADZU Shim-pack VP-ODS C18在柱温为28℃,流动相为乙腈∶甲醇∶水(磷酸调pH 2.5)为70∶10∶20(v/v/v),流速为1 mL/min,进样量20μL,荧光检测波长为λex=331 nm和λem=500 nm条件下,桔霉素在0.05~5 mg/L范围内线性关系良好,最低检测限为5μg/L,在红曲霉发酵液中平均加标回收率为98.17%,RSD=1.8%。

【Abstract】 Citrinin is a kind of metabolites of Monascus,which is toxic to kidney.In this paper,we established a effective determination method of citrinin content by HPLC,and it’s as follows: The column was Shimadzu Shim-pack VP-ODS C18(250 mm×4.6 mm),the mobile phase composition was V(acetonitrile)∶V(methano1)∶V(water) is 70∶10∶20,pH 2.5.And the column temperature was at 28 ℃,the flow rate was 1.0 mL/min.Then citrinin content was detected by a fluorescence detector at λex=331 nm and λem=500 nm.Under this chromatographic conditions,the concentration of citrinin from 0.05 to 5 mg/L has a good linear relationship.The minimum determination limit was 5 μg/L,the average recovery rate was 98.17%,and RSD was 1.8%.
