人参皂苷Rg1对痴呆大鼠脑匀浆中BMSCs的神经性分化作用

【摘要】 目的研究人参皂苷Rg1对痴呆大鼠脑匀浆中BMSCs的神经性分化作用。方法通过贴壁法分离大鼠骨髓间充质干细胞,体外培养扩增,观察人参皂苷Rg1及痴呆大鼠脑匀浆对其分化影响,光镜下观察细胞形态,免疫细胞化学检测神经元特异性烯醇化酶(NSE)的表达。结果痴呆大鼠脑匀浆中BMSCs的神经性分化作用受到抑制,人参皂苷Rg1加入72h后,神经元样分化明显,免疫细胞化学染色NSE呈阳性。结论加入人参皂苷Rg1后,BMSCs在痴呆大鼠脑组织匀浆液中NSE阳性表达率提高。

【Abstract】 Objective To investigate whether ginsenoside Rg1 can induce rat bone marrow mesenchymal stem cells(BMSCs)to differentiate into neuron-like cells or not in homogenate of dementia rats.Methods BMSCs were isolated by wall sticking method,amplifying cultured in vitro,and differentiated into neuron-like cells by inducement with ginsenoside Rg1and homogenate of dementia rats.The morphology of cells was observed under light microscopy,and neuron-specific enolase(NSE)were detected by immunocytochemistry. Results Differentiation of BMSCs in homogenate of dementia rats was depressed.After being induced by ginsenoside Rg1 for 72 hours,some of the cells would be induced into neuron-like cells(15.36%±2.35%),showing positive of NSE by immunochemical staining.Conclusions NSE positive cells of rats bone marrow mesenchymal stem cells can be increased by ginsenoside Rg1 in homogenate of dementia rats
