银杏内酯、银杏黄酮对脑淋巴阻滞后SAH大鼠脑组织HO-1表达的影响

【摘要】 目的探讨银杏内酯、银杏黄酮对脑淋巴引流阻滞(CLB)后蛛网膜下腔出血(SAH)大鼠脑组织血红素氧合酶-1(HO-1)表达的影响。方法将48只Wistar大鼠分为正常对照组、SAH组、SAH+CLB组、SAH+CLB+生理盐水(NS)组、SAH+CLB+银杏内酯干预组(又分为低剂量、高剂量组)、SAH+CLB+银杏黄酮干预组(又分为低剂量、高剂量组)。用结扎颈部淋巴管、摘除淋巴结及注入自体动脉血法制作CLB和SAH大鼠模型。制作CLB模型后24h诱导SAH模型。用RT-PCR法检测大鼠脑组织HO-1mRNA的表达。结果各模型组大鼠脑组织HO-1mRNA表达明显高于正常对照组(均P<0.01),其中以SAH+CLB组、NS组表达进一步增强。银杏内酯和银杏黄酮组HO-1mRNA的表达水平明显高于SAH+CLB组、NS组(均P<0.01),其中两高剂量组表达又明显高于低剂量组(均P<0.01),两低剂量组大鼠脑组织HO-1mRNA的表达与SAH+CLB组及NS组差异无统计学意义(P>0.05)。结论 CLB可诱导SAH鼠脑组织HO-1表达水平明显增高,银杏内酯和银杏黄酮对之有促进作用,尤以高剂量组效果更明显。

【Abstract】 Objective To investigate the effects of ginkgolide and ginkgetin on expression of heme oxygenase-1(HO-1)in cerebral tissue following subarachnoid hemorrhage(SAH)and blockade of cerebral lymphatic drainage(CLB).Methods The 48 Wistar rats were divided into normal control,SAH group,SAH+CLB group,SAH+CLB+NS group,higher and lower dose of ginkgolide groups,higher and lower dose of ginkgetin groups.One day after the induction of CLB model,SAH model was produced.The expression of HO-1 mRNA in the brain were determined by RT-PCR.Results The expression of HO-1 mRNA in rat brain of the model group was significantly higher than the normal control group(all P<0.01).The expression of SAH+CLB and NS group was more higher.The expression level of HO-1 mRNA in brain of ginkgolides and ginkgetin higher-dose group was significantly higher than the SAH+CLB group and NS group(P<0.01).The expression of two high-dose group also was significantly higher than that of low-dose groups(all P<0.01).Conclusion HO-1 expression level of brain tissue was significantly increased after SAH+CLB,Ginkgo ginkgolides and ginkgetin could enhance the expression of HO-1mRNA.Particularly,the effect of high-dose group was more obvious.
