毛细管区带电泳法测定三白草中芦丁和槲皮素的含量

【摘要】 目的:建立毛细管区带电泳法测定三白草中芦丁和槲皮素的含量。方法:采用石英毛细管(50 cm×50μm i.d.,有效长度为41 cm),以15 mmol/L硼砂(pH 8.5)为背景电解质,运行电压28 kV,重力进样10 cm,12 s,紫外检测波长254nm。结果:芦丁在0.5~100 mg/L浓度范围内线性关系良好(r=0.999 3),平均回收率为97.2%。槲皮素在2.0~200mg/L浓度范围内线性关系良好(r=0.999 6),平均回收率为96.0%。结论:该法简便、快速、污染小,适用于三白草药材中芦丁和槲皮素含量的测定。

【Abstract】 AIM:To establish a capillary zone electrophoresis method for determining contents of rutin and quercetin in Saururus Chinensis(Lour) Bail.METHODS:All experiments were carried out in a fused silica capillary(50 cm×50 μm i.d.,41 cm to the detector) by using 15 mmoL/L borax solution(pH 8.5) as the background electrolyte at a running voltage of 28.0 kV.Samples were injected by a gravity injection method with 10 cm hydrostatic pressure for 12 s.UV detection was performed at 254 nm.RESULTS:The calibration curves was linear in the range of 0.5~100 mg/L for rutin(r=0.999 3) and 2.0~200 mg/L for quercetin(r=0.999 6),respectively.The average recovery of rutin was 97.2% and that of quercetin was 96.0%.CONCLUSION:The method is rapid,simple and produces low pollution.It can be used for measuring the contents of rutin and quercetin in Saururus Chinensis(Lour) Bail.
