维生素A缺乏对大鼠十二指肠DMT1,FPN1 mRNA表达的影响
EFFECT OF VITAMIN A DEFICIENCY ON DMT1,FPN1 mRNA EXPRESSION IN THE DUODENUM IN RATS
【摘要】 目的研究维生素A(VA)缺乏对大鼠十二指肠二价金属离子转运蛋白(DMT1),膜铁转运蛋白(FPN1)mRNA表达的影响,为进一步研究维生素A缺乏影响铁代谢的相关机制提供基础。方法雄性SD大鼠52只,按体重随机分为4组,每组13只,Ⅰ组VA正常对照组,Ⅱ组VA完全缺乏组,Ⅲ组VA轻度缺乏组,IV组VA和铁轻度缺乏组。实验动物喂饲8w后处死,测定血清VA、血红蛋白、血清铁、血清铁蛋白,并用逆转录—聚合酶链反应(RT—PCR)法检测各组大鼠十二指肠二价金属离子转运蛋白(DMT1),膜铁转运蛋白(FPN1)mRNA表达的情况。结果 VA缺乏可使血清铁、血清铁蛋白含量显著降低。VA缺乏时十二指肠二价金属离子转运蛋白(DMT1)、膜铁转运蛋白(FPN1)mRNA的表达显著增强。结论 VA缺乏可能通过多种途径使大鼠十二指肠DMT1、FPN1 mRNA的表达增强,需要进一步研究证实。
【Abstract】 Objective To study the effect of vitamin A(VA) deficiency on the level of divalent metal transporter 1 and ferroportin 1 mRNA expression in the duodenum and to provide basis for the further study on the related mechanism about the effect of vitamin A deficiency on iron metabolism.Method Fifty two male SD rats were divided randomly into four groups according to their body weights and every group included 13 rats.They were Ⅰnormal VA group,Ⅱ entire VA deficient group,Ⅲ slight VA deficient group and Ⅳ slight VA and iron deficient group.After they were raised for 8 w,the rats were sacrificed and the serum VA,hemoglobin,serum iron,serum ferritin were determined.The level of divalent metal transporter 1 and ferroportin 1 mRNA expression in the duodenum were measured using reverse transcription polymerase chain reaction(RT-PCR) method.Results Serum iron and ferritin were decreased(P< 0.05).but the level of divalent metal transporter 1 and ferroportin 1 mRNA expression in the duodenum were incresed significantly(P < 0.05) in VA deficiency.Conclusion VA deficiency may enhance the level of divalent metal transporter 1 and ferroportin 1 mRNA expression in duodenum and further study is needed to comfirm.
