2型老年糖尿病患者外周血内皮祖细胞黏附功能与sCD44s黏附分子临床意义研究

【摘要】 目的观察2型糖尿病(T2DM)老年患者外周血内皮祖细胞(endothelial progenitor cells,EPCs)黏附功能和sCD44s变化,探讨二者在T2DM发生发展中的作用。方法选择老年T2DM患者20例和正常对照10例,密度梯度离心法从外周血获取单个核细胞,接种在培养板内,培养7d后对贴壁细胞进行测定。流式细胞仪鉴定FITC标记荆豆凝血素Ⅰ(FITC-UEA-I)和DiI标记的乙酰化低密度脂蛋白(DiI-acLDL)双阳性细胞为正常分化的EPCs。胰蛋白酶消化后计数贴壁细胞观察EPCs黏附能力。采用定量ELISA法测定10例老年糖尿病患者血清中sCD44s的含量。结果①T2DM患者外周血EPCs黏附细胞数较正常对照组显著减少(P<0.01);T2DM患者随血糖、糖化血红蛋白升高,EPCs黏附细胞数逐渐降低,两组间有统计学差异(P<0.05)。T2DM患者外周血sCD44s较正常对照组升高(P<0.05);T2DM患者随血糖、糖化血红蛋白升高,sCD44s逐渐升高,两组间无统计学差异(P>0.05)。②T2DM患者EPCs黏附能力与sCD44s二者间相关系数r=0.361(P>0.05),二者间无相关性。结论 T2DM患者外周血EPCs的黏附能力明显受损,但与血清中sCD44s的升高无相关性,EPCs的生物学功能仍需进一步研究。

【Abstract】 Objective To investigate the function of endothelial progenitor cells(EPCs) from peripheral blood of the older 2-DM patient(T2DM) and study the role of EPCs in the development of the T2DM patient.Methods EPCs isolated from human peripheral blood of the older T2DM patient(n=20) and age matched control subjects(n=10).Cytochemical analysis was conducted after 7 days culture.EPCs were characterized as Dil-acLDL/FITC-UEA-1 double positive cell detected by FCM.EPCs adhesion assay was performed by replating those on fibronectin-coated dishes,and then adherent cells were counted.The sCD44s were detected by ELISA in 10 cases of DM.Resuts 1.The functional adhesions of EPCs were significantly impaired in the older T2DM patient.2.The sCD44s were rised significantly with the increase of the glouse and HbAlc.There were not corrlation between the adhesion of EPCs and the sCD44s.Conclusion These findings suggest EPCs may be play an important role in the development of DM.EPCs dysfunction need be further investigated.
