银耳多糖的抗肿瘤作用及其机制

【摘要】 目的观察银耳多糖对荷H22肝癌小鼠的抑瘤作用,应用基因表达谱芯片研究其抗肿瘤作用的机制。方法建立荷H22肝癌的小鼠模型,用肿瘤抑制率评价银耳多糖对肿瘤的抑制作用;用基因表达谱芯片研究其抗肿瘤作用的机制。结果银耳多糖在6 mg.kg-1时抑制肿瘤效果最好,抑瘤率为72.3%。基因表达谱芯片分析结果表明,共有324个基因有表达差异。其中表达上调基因185个,表达下调基因139个。经初步分析,主要为信号转导基因、DNA损伤检测/p53和ATM通路基因、抗原递呈基因、化疗应答相关基因等。结论银耳多糖对肿瘤有较好的抑制作用,其抗肿瘤作用是多靶点、多因素作用的结果,既与癌症相关基因有关,又与免疫调节、信号传导等基因有关。

【Abstract】 Objective To investigate the inhibition effect on H22 hepatoma by polysaccharides from Tremella fuciformis and explore the mechanism by applying gene expression array chip. Methods The H22 hepatoma model was set up in mice.The inhibition rate was used to evaluate anti-tumor effects of polysaccharide from Tremella fuciformis and tumor inhibitory mechanism was explored by using cDNA microarray. Results The best inhibition rate was showed as 72.3%(P<0.01) by Tremella fuciformis polysaccharides at the dose of 6 mg·kg-1.There was expression difference in 324 of the total 4 000 genes compared with negative control groups,185 genes of which were up-regulated and 139 genes were down regulated.By the prelinminary analysis,most of genes were those about signal conduction,DNA injury check/P53 and ATM path,antigen presentation and chemotherapy response. Conclusion The polysaccharide from Tremella fuciformis inhibits H22 hepatoma in mice markedly.The mechanism of action is multitargeted and multiple factors involved,which linking either to cancer associated gene or to immune regulation and signal conduction.
