环孢霉素A联合干扰素-α对膀胱癌T24/ADM耐药逆转的研究
Study of Cyclosporin A combined with INF-α to revert the multidrug resistance of bladder cancer T24/ADM cell line
【摘要】 目的:探讨逆转人膀胱癌T24/ADM多药耐药(MDR)的可行性。方法:采用MTT比色法研究单独逆转组[环孢霉素A(CsA)或干扰素-α(INF-α)]和联合逆转组(CsA+INF-α)对阿霉素(ADM)细胞毒性在人膀胱癌T24细胞的影响。结果:CsA与INF-α能部分逆转膀胱癌T24细胞对ADM的敏感性。联合两药比单独使用一种有更高的细胞抑制作用,当CsA浓度为1.0μg/ml、INF-α为2.0×106 U/L时,细胞抑制率达44.7%(P<0.01)。结论:CsA、INF-α能逆转膀胱癌T24/ADM的耐药性,联合使用比单独使用一种有更好的效果。
【Abstract】 Objective: To probe into the possibility of reversion of the multidrug resistance(MDR) in bladder cancer cell line T24.Methods: The effect of alone group [Cyclosporin A(CsA) or INF-α] and combination group(CsA+INF-α) on the cytotoxicity of Adriamycin(ADM) in human bladder cancer cell line T24 in vitro were studied in vitro by MTT colorimetric assay.Results: The results showed that drug sensitivty to ADM were partly reestablished by CsA or INF-α.The apoptosis rate in combination group of CsA combined with INF-α was higher than that in either of the two alone,the dose of CsA was 1.0 μg/ml,INF-α was 2.0×106 U/L,the inhibition rate was 44.7%(P<0.01).Conclusion: CsA and INF-α can revert the multidrug resistance of bladder cancer cell line T24.Combination of CsA and INF-α have better result than either of the two alone.
