配制网织红细胞染色液的改进

【摘要】 目的网织红细胞(reticulocyte,Ret)是判断骨髓造血功能的重要指标,然而网织红细胞染液的配制及其染色过程比较繁琐,为弥补以上不足,受血球计数仪稀释液启发,我们改良了常规所用的煌焦油蓝生理盐水染液。在保证同等质量的前提下,减少染液配制的繁琐过程,延长其保存时间,提高染色质量等。方法选取增生性贫血患者外周血,分别用改良法和常规法在不同时间,不同温度下染色,然后推片计数。结果改良法染色效果更佳,杂质和细胞碎片明显减少,时间对改良法染色结果影响较小。结论在保证同等质量前提下,改良法染色网织红细胞,减少了染色液配制的繁琐,延长了保存时间,细胞碎片、杂质均少于常规法,同时降低了染色对时间的要求。

【Abstract】 Objective Reticulocyte is an important indicator to determine the bone marrow function,but it’s complicated to compound biological stain and staining the cell.In order to make up this shortage,be inspired by the liquefier of corpuscl count instrument,we improved the traditional Brilliant cresyl bulue.This kind of dyestuff can reduce the process of preparation,extend guarantee period of dye and improve dyeing quality.Methods Dyeing the peripheral blood from patients who with proliferative anemia in different way.In different temperature,one group be stained by the original method,at the same time we stained another group by the new material.Then we can make the slide and count reticulocyte after a period time.Results Improved staining is better than the old one.For example,impurities and cell debris was significantly reduced,improved method of staining was less affected by the using time.Conclusion With the same quality,new staining material can prolong the period of validity,reduce the complicate process of compound the coloring agent,significantly reduced impurities and cell debris and depress the definition of the cost time.
