六神丸和雄黄中砷的组织分布研究

【摘要】 本文研究Beagle犬服用不同剂量雄黄和六神丸并经过28天服药期和14天恢复期后,组织中总砷及各形态砷化合物的分布情况,为六神丸的安全性评价提供依据。采用ICP-MS测定Beagle犬灌服不同剂量雄黄及六神丸后各组织中总砷浓度,HPLC-ICP-MS测定各组织中砷的形态分布。随着雄黄的服用剂量增大,Beagle犬组织中总砷含量和亚砷酸盐(AsIII)+二甲基砷酸(DMA)浓度增大。含相同剂量雄黄的六神丸组与雄黄组相比,多数组织中总砷含量降低,所有组织中的AsIII+DMA浓度均降低,而肝、脾、肾中的砷甜菜碱(AsB)浓度升高。总砷含量在组织中呈现出一定的剂量依赖关系,六神丸复方配伍后对雄黄具有一定的减毒作用。

【Abstract】 This study is to report the tissue distribution of arsenic after giving different doses of realgar and Liushen pills to Beagle dogs,in order to provide basis for the safety evaluation of Liushen pills.ICP-MS was used to measure arsenic concentration,and HPLC-ICP-MS was used to analyze arsenic speciation.The concentration of total arsenic and AsIII + DMA(arsenite + dimethylarsenic acid) increased with dosing of realgar.Total arsenic concentration in most tissues and AsIII + DMA concentration in all tissues of Liushen pills group are lower than that of realgar group,but AsB concentration in liver,spleen and kidney of Liushen pills group increased.The concentration of total arsenic showed a dose-dependent manner with dosage administered.It was indicated that components in Liushen pills can reduce solubility of arsenic in realgar,which may decrease toxicity of realgar.
