蛹虫草Fjnu-01高产虫草素的液体培养基优化
The optimization of the culture media for high production of cordycepin by Cordyceps militaris Fjnu-01
【摘要】 优化液体培养蛹虫草(Cordyceps militaris)Fjnu-01菌株培养条件,用高效液相为检测手段,以发酵液中虫草素的含量作为指标,通过单因子实验和均匀设计对蛹虫草产虫草素的液体发酵培养基配方进行优化。结果显示,蛹虫草Fjnu-01菌株产虫草素的最适碳源为蔗糖+玉米粉,最适合氮源为蛋白胨,最适生长因子为VB1,最适pH为6.4。最适培养基配方为:蔗糖2.80%;玉米粉0.54%;蛋白胨1.91%;KH2PO4 0.28%;MgSO4 0.15%;VB1 0.11‰。
【Abstract】 In the present paper,the culture condition for producing cordycepin by Cordyceps militaris Fjnu-01 was optimized via single factor experiments and uniform design.The content of cordycepin was assayed by HPLC,and the results showed that the optimal carbon source,nitrogen source,growth factor and optimal pH value were sucrose and corn flour,peptone,VB1 and pH value of 6.4,respectively.The optimal medium concentration was shown as follows: sucrose 2.80%,corn flour 0.54%,peptone 1.91%,KH2PO4 0.28%,MgSO4 0.15% and VB1 0.11‰.
