尼古丁对人牙周膜细胞基质金属蛋白酶-1、3表达的影响
Effects of nicotine on the production of MMP-1,3 in human periodontal ligament cells
【摘要】 目的:探讨尼古丁对体外培养人牙周膜细胞(hPDLCs)基质金属蛋白酶-1、3(MMP-1、3)表达的影响及可能机制。方法:采用原代组织块法培养hPDLCs,应用逆转录多聚酶链反应(RT-PCR)及酶联免疫吸附测定法(ELISA)检测尼古丁单独及联合应用α7亚型乙酰胆碱受体特异性拮抗剂α-银环蛇毒素(α-BTX)作用下,hPDLCs中MMP-1、3表达的变化。结果:尼古丁(10-4mol/L)作用于hPDLCs,其MMP-1、3mRNA及蛋白表达均有升高(P<0.05),此作用可被α-BTX(1.25×10-9mol/L)拮抗。结论:尼古丁可使体外培养的hPDLCs中MMP-1、3表达升高,此过程可能由α7nAChR通路介导。
【Abstract】 AIM: To observe the effects of nicotine and/or α-BTX on the production of MMP-1 and 3 in human periodontal ligament cells cultured in vitro.METHODS: Human periodontal ligament cells were cultured using explant culture technique.The cells were put in the presence of nicotine and/or α-BTX,a specific angatonist of α7-nicotinic aeetylcholine receptor(α7nAChR),at the concentration of 10-4mol/L and 1.25×10-9mol/L,respectively.Then the expression of MMP-1 and 3 was measured by reverse transcription-polymerase chain reaction(RT-PCR) and enzyme linked immunosorbent assay(ELISA).RESULTS: At the exposure of nicotine alone,MMP-1 and 3 were both increased significantly at the mRNA and protein level,which ould be inhibited by α-BTX.CONCLUSION: Nicotine can stimulate MMP-1 and 3 production by hPDLCs and this process can be mediated by α7nAChR pathway.
