IL-24抑制宫颈癌裸鼠移植瘤生长的初步研究
A Preliminary Study on Inhibitory Effect of IL-24 in Nude Mice Model of Cervical cancer
【摘要】 目的探讨基因重组质粒pDC316-hIL-24对裸鼠人宫颈癌移植瘤生长和浸润转移的抑制作用,以及其与nm23H1表达变化的关系。方法将18只造模成功的人宫颈癌Hela细胞裸鼠模型随机分为3组,每组6只:BPS缓冲液对照Ⅰ组,空质粒Ⅱ组,IL-24重组质粒Ⅲ组。观察各组裸鼠饮食、体重等一般状况,记录各组裸鼠移植瘤体积、绘制生长曲线。观察裸鼠肿瘤附近淋巴结转移情况,实验结束后根据瘤体重量计算肿瘤抑制率。应用RT-PCR法检测移植瘤IL-24的表达,并用免疫组化法nm23H1蛋白表达变化。结果基因重组质粒pDC316-hIL-24在裸鼠体内转染成功,且有明显的抑瘤效果,抑瘤率为42.9%,与对照组、空质粒组差异有统计学意义(P<0.05)。IL-24重组质粒组肿瘤淋巴结转移较其他组少,差异有显著性(P<0.05)。免疫组化结果显示,IL-24组的转移抑制基因nm23H1蛋白表达明显增多,与其余2组比较差异有统计学意义(P<0.001)。结论重组质粒pDC316-hIL-24能明显抑制宫颈癌裸鼠肿瘤模型的生长,并可能通过上调nm23H1的表达发挥抗肿瘤生长和浸润转移的作用。
【Abstract】 Objective To study he inhibition function of gene recombinant plasmid pDC316-hIL-24 on human cervical cancer nude mice model tumor growth and invasion.Method Nude mice xenograft models were established with human cervical cancer Hela cells,and then animal models were randomly divided into three groups,each group had 6 mice: PBS(groupⅠ),empty plasmids(group Ⅱ),recombinant plasmids pDC316-hIL-24(group Ⅲ).All the nude mice were injected into the tumor-bearing mice with Lipofectamine 2000 to interfere with the growth of transplanted tumor.Their effects on the volume of the tumor and inhibitory rate of the mice were observed.hIL-24 expression was determined in transplanted tumor by PCR.Lymph node metastasis was observed,and as well as immunohistochemistry was employed for investigating nm23H1 albumen in each group.Results ①After transfection IL-24 gene was successfully transfected into and expressed in tumor cells.Its function of inhibiting tumor growth was 42.9%,significantly higher than PBS,empty plasmids(P<0.05).②Lymph node metastasis was decreased in group Ⅲ(P<0.05).Immunohistochemistry showed that,the expression of nm23H1 albumen in IL-24 group was significantly higher than other two groups(P<0.05).Conclusion Gene recombinant plasmid pDC316-hIL-24 can significantly inhibit the growth of transplanted human cervical cancer in nude mice.IL-24 may activate expression of nm23H1 albumen to inhibit human cervical cancer nude mice model tumor growth and invasion.
