薏苡仁提取液对AOM诱发结肠癌大鼠模型组织病理学变化的影响

【摘要】 目的观察薏苡仁提取液对氧化偶氮甲烷(AOM)所致结肠癌大鼠模型组织病理学变化的影响。方法随机将20只3周龄断乳雄性SD大鼠分为阴性对照组、阳性对照组、实验组,其中实验照组给予不同浓度的薏苡仁提取液喂养,时间为AOM诱导前、后各5周,共10周。阳性对照组和实验组大鼠每周腹腔注射AOM 20ms/kg,持续2周,造成大鼠结肠癌模型。各组均于10周后取结肠组织,用免疫组织化学方法观察大鼠结肠癌组织细胞内环氧化酶(COX2)、癌胚抗原(CEA)蛋白的表达,并进行图像分析。结果阴性对照组大鼠的结肠组织内未见癌胚抗原蛋白表达,而阳性对照组和实验组的CEA均呈阳性表达,阳性产物呈深棕色,定位于细胞核中,细胞质内仅有少量表达。随着实验组给予薏苡仁提取液浓度的提高,CEA、COX2阳性表达逐渐下降,组间差异有统计学意义(P<0.05)。结论薏苡仁提取液可下调AOM诱导结肠癌大鼠COX2、CEA蛋白的表达,在结肠癌的防治中具有一定的作用。
【Abstract】 Objective To observe the effect of Coixenolide Extract on the Histopathological changes induce by zoxymethane(AOM) in rat colon cancer model.Methods Twenty 3 week-old SD male rats were randomly divided into three groups Negative control group,positive control group,experimental control group,the experimental control group was given Coixenolide Extract before and after AOM induced colon cancer.To induce rat colon cancer,3 weeks-old SD male rats received ip injections of AOM at a dose of 20mg/kg once a week for 10 weeks.Immunohistochemical method was used to detecte the expression of protein COX2 and CEA in the rat colon tissue and image analysis was used to analyze the immunoreactivity.Result No expression of the protein COX2 and CEA were found in the normal rat colon tissue,but a significant increase was seen in the rat colon tissue in the following groups: experimental control group and the positive control group,and significant difference was found between each group(P<0.05).Deep brown color of immunoreactivity was found to localize in the nucleus,and the expression was much in the cytoplasm less.Conclusion Coixenolide extract can be reduced expression of COX2 and CEA in colon cancer in rats.
