胃癌组织中KiSS-1和基质金属蛋白酶-9蛋白及mRNA的表达
Expressions of KiSS-1 and MMP-9 protein and mRNA in gastric cancer tissue
【摘要】 目的探讨肿瘤转移抑制基因KiSS-1及基质金属蛋白酶9(MMP-9)与胃癌侵袭、转移的关系。方法采用免疫组织化学SP法和逆转录聚合酶链反应(RT-PCR)检测40例胃癌及40例正常胃黏膜组织中KiSS-1及MMP-9蛋白和mRNA的表达情况,分析其与胃癌患者各临床病理特征的关系及二者的相关性。结果胃癌组织中KiSS-1蛋白的阳性表达率(52.5%)低于正常胃黏膜组织(95.0%)(P<0.05),并且其低表达与肿瘤浸润深度和淋巴结转移密切相关(P<0.05)。胃癌组织中MMP-9蛋白的阳性表达率(77.5%)高于正常胃黏膜组织(52.5%)(P<0.05),并且其高表达与肿瘤浸润深度和淋巴结转移相关(P<0.05)。胃癌组织中KiSS-1 mRNA的阳性表达率及表达水平(57.5%,0.869±0.063)均低于正常胃黏膜组织(92.5%,1.103±0.152)(P<0.05),并且其低表达与肿瘤浸润深度和淋巴结转移密切相关(P<0.05)。胃癌组织中MMP-9 mRNA的阳性表达率及表达水平(75.0%,1.083±0.137)均高于正常胃黏膜组织(47.5%,0.902±0.035)(P<0.05),并且其高表达与肿瘤浸润深度和淋巴结转移密切相关(P<0.05)。KISS-1与MMP-9蛋白和mRNA的表达均呈负相关(P<0.05)。结论 KiSS-1的低表达和MMP-9的过表达可能与胃癌的浸润、转移有关。二者有望成为判定胃癌侵袭和转移能力的指标。
【Abstract】 【Objective】 To explore the relationship between metastasis suppressor gene KISS-1,matrix metalloproteinase-9(MMP-9) and the invasion,metastasis of gastric cancer.【Methods】 The expressions of KiSS-1 and MMP-9 protein and mRNA in specimens taken from 40 cases of gastric cancer and 40 cases of normal gastric mucosa tissue were detected by immunohistochemicalassay(SP) and reverse transcription-polymerase chain reaction(RT-PCR).To analyze the relationship between them and clinical data.【Results】 The positive expression rate of KiSS-1 protein in gastric cancer(52.5%) was significantly lower than that in normal gastric mucosa tissue(95.0%)(P<0.05).The low expression of KiSS-1 protein in gastric cancer was related with the depth of invasion and lymph node metastasis(P<0.05).The positive expression rate of MMP-9 protein in gastric cancer(77.5%) was significantly higher than that in normal gastric mucosa tissue(52.5%)(P<0.05).The high expression of MMP-9 protein in gastric cancer was related with the depth of invasion and the metastasis of lymph node(P<0.05).The positive expression rate and relative contents of KiSS-1 mRNA in gastric cancer(57.5%,0.869±0.063) were significantly lower than those in normal gastric mucosa tissue(92.5%,1.103±0.152)(P<0.05).The low expression of KiSS-1 mRNA in gastric cancer was related with the depth of invasion and lymph node metastasis(P<0.05).The positive expression rate and relative contents of MMP-9 mRNA in gastric cancer(75.0%,1.083±0.137) were significantly higher than those in normal gastric mucosa tissue(47.5%,0.902±0.035)(P<0.05).The high expression of MMP-9 mRNA in gastric cancer was related with the depth of invasion and the metastasis of lymph node(P<0.05).The expression of KiSS-1 had negative correlation with the expression of MMP-9(P<0.05).【Conclusion】 Low expression of KiSS-1 and high expression of MMP-9 maybe related with invasion and metastasis of gastric cancer.KiSS-1 and MMP-9 could be a marker to evaluate invasion and metastasis in gastric cancer.
