【摘要】 目的探讨肿瘤转移抑制基因KiSS-1及基质金属蛋白酶9(MMP-9)与胃癌侵袭、转移的关系。方法采用免疫组织化学SP法和逆转录聚合酶链反应(RT-PCR)检测40例胃癌及40例正常胃黏膜组织中KiSS-1及MMP-9蛋白和mRNA的表达情况,分析其与胃癌患者各临床病理特征的关系及二者的相关性。结果胃癌组织中KiSS-1蛋白的阳性表达率(52.5%)低于正常胃黏膜组织(95.0%)(P<0.05),并且其低表达与肿瘤浸润深度和淋巴结转移密切相关(P<0.05)。胃癌组织中MMP-9蛋白的阳性表达率(77.5%)高于正常胃黏膜组织(52.5%)(P<0.05),并且其高表达与肿瘤浸润深度和淋巴结转移相关(P<0.05)。胃癌组织中KiSS-1 mRNA的阳性表达率及表达水平(57.5%,0.869±0.063)均低于正常胃黏膜组织(92.5%,1.103±0.152)(P<0.05),并且其低表达与肿瘤浸润深度和淋巴结转移密切相关(P<0.05)。胃癌组织中MMP-9 mRNA的阳性表达率及表达水平(75.0%,1.083±0.137)均高于正常胃黏膜组织(47.5%,0.902±0.035)(P<0.05),并且其高表达与肿瘤浸润深度和淋巴结转移密切相关(P<0.05)。KISS-1与MMP-9蛋白和mRNA的表达均呈负相关(P<0.05)。结论 KiSS-1的低表达和MMP-9的过表达可能与胃癌的浸润、转移有关。二者有望成为判定胃癌侵袭和转移能力的指标。更多还原
