miR-383增强SLE患者外周血单个核细胞Foxp3 mRNA的表达

【摘要】 目的 初步探讨microRNA-383(miR-383)对IL-2信号介导的系统性红斑狼疮(SLE)患者外周血单个核细胞(PBMCs)中Foxp3 mRNA转录的影响。方法 采用Western blot方法检测14例SLE患者PBMCs中SOCS3蛋白表达水平,通过相关microRNA软件预测和Western blot鉴定靶向SOCS3的microRNA,RT-PCR方法检测miR-383转染SLE患者的PBMCs中转录因子Foxp3 mRNA的转录水平。结果 Western blot结果发现,SLE患者的PBMCs中SOCS3蛋白表达水平显著增加,miR-383抑制SOCS3蛋白表达。SLE患者PBMCs转染miR-383后Foxp3 mRNA的转录水平显著增高。结论 miR-383靶向抑制SOCS3,上调IL-2信号介导的SLE患者的PBMCs中Foxp3 mRNA的转录水平。

【Abstract】 Objective To primarily explore the role of microRNA-383(miR-383) in modulation of IL-2-mediated Foxp3 mRNA transcription in systemic lupus erythematosus(SLE) patients’ peripheral blood mononuclear cells(PBMCs).Methods Western blot assay was performed to measure the expression of SOCS3 protein in 14 SLE patients’ PBMCs.MicroRNA of target for SOCS3 was miR-383 by forecasting of relative microRNA softwares and then by Western blot.Moreover,RT-PCR assay was used to test the effect of miR-383 on Foxp3 mRNA transcription level in SLE patients’ PBMC cells.Results Western blot assay showed increasing expression of SOCS3 protein in 14 SLE patients’ PBMCs in different extent.Western blot assay verified that miR-383 could inhibit the expression of SOCS3 protein.RTPCR assay exhibited that miR-383 could increase Foxp3 mRNA transcriptional level in SLE patients’ PBMCs.Conclusion miR-383 promotes IL-2-mediated Foxp3 mRNA transcription by target for SOCS3 in SLE patients’ PBMCs.
