RNAi技术阻抑CTGF对瘢痕疙瘩形成影响的体外研究

【摘要】 目的 用RNA干扰(RNAi)技术探讨结缔组织生长因子(CTGF)对瘢痕疙瘩中胶原代谢的影响。方法采用RNAi技术,将针对人CTGF基因设计合成的3对特异性小干扰RNA(siRNA)-CTGF分别转染体外培养的人瘢痕疙瘩成纤维细胞(KFB),通过逆转录-多聚酶链反应(RT-PCR)和Western blot检测转染后瘢痕疙瘩组织中CTGF表达量的变化及胶原含量的变化,并与对照组比较。结果 第3对(C3)siRNA-CTGF转染人KFB后CTGF的基因表达及胶原蛋白表达水平显著受抑,其抑制率分别为86.8%及65.6%。结论 质粒siRA-CTGF转染人KFB后有效沉默CTGF的表达进而使胶原合成降低,CTGF对瘢痕疙瘩形成过程中胶原蛋白的合成有促进作用。

【Abstract】 Objective To investigate the effect of connective tissue growth factor(CTGF) on the collagen metabolism in human keloid fibroblasts(KFB) with RNA interference(RNAi).Methods With the application of RNAi,human KFB were respectively transfected in vitro by 3 pairs of specific small interfering RNA(siRNA)-CTGF gene synthesized for human CTGF to detect the variations of expression level and collagen c ontent of CTGF in transfected keloid fibroblasts by reverse transcription polymerase chain reaction(RT-PCR) and Western blotting to compare with the control group.Results The expression of the genes and proteins of CTGF was remarkably inhibited following the transfection of human KFB by the 3rd pair(C3) siRNA-CTGF,with inhibition rates of 86.8% and 65.6%.Conclusion Human KFB transfec ted by plasmid siRNA-CTGF can effectively silence the expression of CTGF,thus reducing collagen synthcsis.CTGF can promote the collagen synthesis in keliod development.
