参麦及缺血预处理对大鼠肝缺血再灌注损伤的保护作用

【摘要】 目的:研究参麦注射液及缺血预处理对肝缺血再灌注损伤的保护作用。方法:64只Wistar大鼠随机分成4组:假手术组(sham-operation,SO);缺血再灌注组(ischemia reperfusion,IR);缺血预处理组(ischemic preconditioning,IPC);参麦组(Shengmai in-jection,SM)。建立肝脏缺血再灌注损伤模型。SO组行剖腹假手术;IR、IPC、SM组于再灌注60 min、90 min时分别随机取8只大鼠检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、乳酸脱氢酶(LDH)和白细胞介素6(IL-6),同时取肝组织制成匀浆检测金属硫蛋白(MT),SO组于相应时点取样检测相同指标。分析比较各组计量资料。结果:SM组及IPC组在再灌注60 min、90 min时ALT、AST、LDH、IL-6水平均低于IR组(P<0.01),而金属硫蛋白水平均高于IR组。结论:参麦及缺血预处理对肝缺血再灌注损伤具有保护作用。

【Abstract】 Objective:To investigate the protective effect of Shengmai injection and ischemic preconditioning(IPC) on liver ischemia reperfusion injury in rat model and to explore its possible mechanism.Methods:Sixty-four healthy male Wistar rats were randomized into four groups,group A(sham-operation,SO),group B(ischemia reperfusion,IR),group C(ischemic preconditioning,IPC) and group D(Shengmai injection,SM).The rat model of ischemia reperfusion injury was established.The group SO received only sham operation.8 rats were randomly taken from the group IR,IPC and SM at 60 min and 90 min after liver reperfusion and detected the serum levels of alanine aminotransferase(ALT),aspartatic aminotransferase(AST),lactate dehydrogenase(LDH),interleukin 6(IL-6) and metallothionein(MT) level in hepatic tissues was determined.The same indexes were determined in group SO at the same time points.Then the measurement data of the indexes were compared among various groups.Results:At 60 min,90 min after liver reperfusion,the serum levels of ALT,AST,LDH and IL-6 in the group SM and IPC were significantly lower than those in the group IR(P<0.01),but the MT level of hepatic tissues in group IPC and SM was markedly higher than that in the group IR(P<0.01).Conclusion:Shengmai and IPC have the protective effect on rat liver ischemia-reperfusion injury.
