甲醛吸入急性肺水肿大鼠出凝血时间变化及地塞米松干预作用
Changes of bleeding time,coagulation time and intervention of dexamethasone on acute pulmonary edema rats induced by inhaling formaldehyde
【摘要】 目的:探讨甲醛吸入急性肺水肿模型大鼠出、凝血时间的变化及地塞米松的干预作用,为其病理生理机制及治疗提供实验依据。方法:大鼠27只随机分为对照组、模型组和地米组,地米组于造模前0.5 h腹腔注射地塞米松1 mg/100 g体质量,其余两组注射等量生理盐水。按文献模型组和地米组以甲醛喷雾吸入制作急性肺水肿模型。染毒7 h后作出血时间和凝血时间测定,8 h后处死,取心、肺等器官称重并计算肺/心系数。对照组以生理盐水替代甲醛,处理方法同模型组。结果:与生理盐水对照组比较,模型组和地米组染毒后8 h体质量均显著降低(均P<0.01),肺湿重显著增大(均P<0.05),肺/心系数增大(P<0.05,P<0.01),出、凝血时间无显著性改变(均P>0.05)。结论:甲醛喷雾吸入造成的急性肺水肿大鼠出、凝血时间变化不明显,地塞米松干预无显著影响。
【Abstract】 Objective:To study the change of bleeding time(BT),coagulation time(CT) and the intervention of dexamethasone on rat model of acute pulmonary edema induced by inhaling formaldehyde.Methods:27 rats were divided into normal saline control group,formaldehyde inhalation(model) group and dexamethasone treated group randomly.Dexamethasone group rats were injected with dexamethasone sodium phosphate injection 1mg/100g by intraperitoneal injection before inhaling formaldehyde vapor,and under the same conditions,other groups were treated by equivalent normal saline.Then the rats of model group and dexamethasone group were induced pulmonary edema by inhaling formaldehyde vapor consecutively for 2 h.BT and CT were determined after 7 h,and the rats were killed 8 h later.Meanwhile,the lungs and heart of rats were cut and weighted,and the cardiopulmonary index and lung index were calculated.Rats of normal saline control group were treated as the same as above,but the normal saline instead of formaldehyde.Results:Compared with normal saline control group,the body weights in both of model and dexamethasone groups were decrease signally(both P<0.01),the cardiopulmonary index of model group(P<0.05) and dexamethasone group(P<0.01) were increase observably,and the lung wet weight was increased significantly in both groups(both P<0.05),but BT and CT showed no significant change(both P>0.05).Conclusion:Bleeding time and coagulation time were not altered significantly after pulmonary edema induced by formaldehyde inhalation and dexamethasone intervention had no more distinct effect on them.
