全蝎纯化液对静脉血栓形成大鼠纤溶和凝血系统的影响
Effects of Scorpion Purified Liquid on Fibrinolysis System and Coagulation System in Rats with Thrombosis
【摘要】 目的通过观察全蝎纯化液对静脉血栓形成大鼠凝血和纤溶系统的影响,探讨全蝎抗静脉血栓形成的作用机制。方法 SD大鼠随机分为空白组、模型组和全蝎纯化液大、中、小剂量组,全蝎纯化液各剂量组由颈外静脉分别注入全蝎纯化液,空白组和模型组注入等体积的生理盐水。采用结扎大鼠腹腔主静脉造成静脉血栓模型,观察其对激活部分凝血活酶时间(APTT)、凝血酶原时间(PT)、凝血酶时间(TT)、组织型纤溶酶原激活物(tPA)、纤溶酶原激活物抑制物-1(PAI-1)等凝血和纤溶指标的影响。结果全蝎纯化液大、中、小剂量能明显减轻血栓重量,与模型组相比,差异有统计学意义(均P<0.01),血栓形成的抑制率分别为75.23%、61.50%、51.05%;能明显延长APTT、PT、TT时间,与模型组相比,差异有统计学意义(均P<0.01);模型组tPA明显降低、PAI-1明显升高,而全蝎纯化液各剂量组能明显抑制PAI-1活性升高、增加tPA活性,与模型组相比,差异有统计学意义(均P<0.01)。结论全蝎纯化液具有明显的抑制血栓形成的作用,其作用机制可能与其抗凝血及促纤溶作用有关。
【Abstract】 Objective To study the effect of scorpion purified liquid on blood coagulation and fibrinolytic system of venous thromboembolism rats and investigate the mechanisms of scorpion purified liquid in preventing venous thrombosis.Methods The SD rats were randomly divided into blank,model and large,medium,small concentration scorpion groups.Three scorpion groups rats were injected scorpion purified liquid through jugular vein,the blank and model groups were injected saline.The rat venous thromboembolism model were established by ligating the main abdominal vein.The indexs of coagulation and fibrinolysis including activated partial thromboplastin time(APTT),prothrombin time(PT),thrombin time(TT),tissue plasminogen activator(t-PA),plasminogen activator inkibitor(PAI) were detected.Results The weigh of thrombosis in large,medium and small concentration groups were significantly lower than model group(P<0.01),and the inhibition rates of venous thrombosis were 75.23%,61.50%,51.05% respectively.APTT,PT,TT were significantly extended compared with model group(P<0.01).The levels of enhance in PAI-1 and reduce in t-PA were significantly inhibited by the scorpion purified liquid compared with model group(P<0.01).Conclusion Scorpion purified liquid has a role on controlling thrombus formation.Its mechanisms of preventing venous thrombosis may be associated with preventing blood from coagulating,promoting the dissolution of fibre.
