新疆产锁阳不同提取部位对Caco-2细胞的抑制作用及其化学成分研究

【摘要】 目的考察锁阳不同提取部位对结肠腺癌细胞系(Caco-2)的细胞抑制活性及其物质基础。方法使用95%乙醇对锁阳全草进行提取,分别用氯仿、乙酸乙酯和正丁醇依次对95%乙醇提取部位进行分段萃取,采用四甲基偶氮唑盐法(MTT)测定各提取部位对体外培养Caco-2的抑制作用,并应用各种色谱技术对氯仿和乙酸乙酯提取部位进行纯化。结果锁阳的氯仿和乙酸乙酯部位具有一定的杀伤Caco-2细胞的作用,IC50分别为88.5μg/ml和179.3μg/ml;从氯仿和乙酸乙酯提取部位分离得到5种化合物,并经1 H-NMR波谱及与相应标准品的薄层色谱比较,确证其结果分别为β-谷甾醇(β-Sitostero,Ⅰ)、熊果酸(ursolic acid,Ⅱ)、胡萝卜苷(β-Sitosterol-3-O-β-glucoside,Ⅲ)、(+)-儿茶素[(+)-catechin,Ⅳ],化合物Ⅴ正在鉴定中。结论锁阳的氯仿和乙酸乙酯提取部位是其主要的抗Caco-2活性部位,可能与从该部位分离到的5种化合物相关。

【Abstract】 Objective To investigate the activities of different extraction parts from the Cynomorium songarium Rupr.in vitro,anti Caco-2 cells and the chemical constituents in contributing to anticanceractivity.Methods The extraction from the Cynomorium songarium Rupr.with 95% ethanol and then from the 95% ethanol extraction part in turn with chloroform,ethyl acetate and n-butanol were conducted.The inhibitory effects of these extraction parts on Caco-2 cells were measured by MTT assay.The chemical constituents in the chloroform and ethyl acetate extraction part were isolated by chromatographys.Results The IC50 of the chloroform and the ethyl acetate extraction part are 88.5 μg/ml and 179.3 μg/ml respectivly;5 componds,β-Sitostero(Ⅰ),ursolic acid(Ⅱ),β-Sitosterol-3-O-β-glucoside(Ⅲ),(+)-catechin(Ⅳ) were isolated from the chloroform and the ethyl acetate extraction part and their structres were verified by 1H-NMR and TLC(compared with the co-rresponding reference substances).Conclusion The chloroform and ethyl acetate extraction part from the Cynomorium songarium Rupr.is the main anti Caco-2 cells active part,which could be associated with the isolated 5 compounds.
